柴苓术芍饮对肝郁脾虚型非酒精性脂肪性肝病大鼠肝组织氧化应激及炎症因子的影响

目的:探讨柴苓术芍饮对肝郁脾虚型非酒精性脂肪性肝病(NAFLD)模型大鼠肝组织氧化应激及炎症因子的影响。方法:选取健康SD大鼠35只,随机分为正常组,模型组,柴苓术芍饮高、中、低剂量组,每组7只。除正常组外,其他4组采用复合因素制备肝郁脾虚型非酒精性脂肪肝模型。成模后给药4周,取肝组织匀浆上清液,采用全自动生化分析仪检测超氧化物歧化酶(SOD)含量;采用酶联免疫吸附法检测丙二醛(MDA)、肿瘤坏死因子(TNF-α)及白介素-1β(IL-1β)的含量;采用蛋白免疫印迹法(WB)测定肿瘤坏死因子(TNF-α)及白介素-1β(IL-1β)蛋白表达。结果:与模型组大鼠比较,柴苓术芍饮高剂量组肝组织SOD活性升高明显,MDA含量、肿瘤坏死因子(TNF-α)及白介素-1β(IL-1β)含量及其蛋白表达降低明显。结论:柴苓术芍饮能改善肝郁脾虚型NAFLD大鼠肝脏氧化应激损伤,降低炎症因子表达。

Objective:To investigate the effects of Chailing Zhushao Decoction on oxidative stress and inflammatory factors in liver tissue of non-alcoholic fatty liver disease(NAFLD)model rats.Methods:35 healthy SD rats were selected and randomly divided into normal group,model group,Chailing Zhoushao Decoction high-dose,medium-dose and low-dose groups,with 7 rats in each group.In addition to the normal group,non-alcoholic fatty liver model of liver depression and spleen deficiency were prepared by compound factors in the other 4 groups.Four weeks after the injection,supernatant of liver tissue homogenate was collected and the contents of superoxide dismutase(SOD)was determined by automatic biochemical analyzer.The contents of malondialdehyde(MDA)、tumor necrosis factor(TNF-α)and interleukin-1β(IL-1β)were detected by ELISA.The protein expressions of tumor necrosis factor(TNF-α)and interleukin-1β(IL-1β)were determined by Western blotting(WB).Results:Compared with model group,SOD activity in liver tissue of Chailing Zhushao Decoction high-dose group was improved significantly,and MDA content、TNF-αand IL-1βcontents and protein expressions were decreased significantly.Conclusion:Chailing Zhushao Decoction can improve liver oxidative stress injury,reduce the expression of inflammatory factors in NAFLD rats with liver depression and spleen deficiency.
