摘要 以超声波辅助法提取野菊花药材中总黄酮,同时使用响应面分析对此方法的提取工艺条件进行优化,主要研究结果如下:1.经单因素试验设计,以野菊花药材中总黄酮含量为考察指标,以影响其五种因素为研究对象,试验结果表明,在乙醇浓度40%,40%浓度的乙醇与原药材的液料比为50∶1(mL/g),超声提取时间为1 h,提取温度为35℃,提取功率为480 W时野菊花药材中总黄酮提取效果最好。2.以前期试验结果为基础,利用响应面分析软件Design Expert,采用Box-Behnken原理进行实验的拟合与分析,并经回归计算发现,39.89%浓度的乙醇与原药材的液料比在50.61∶1(mL/g),超声提取时间为59.83 min,提取温度为35.17℃,提取功率为478.06 W时野菊花药材中总黄酮提取效果最优。 The total flavonoids in wild chrysanthemum herbs were extracted by ultrasonic auxiliary method,and the extraction process conditions of this method were optimized by response surface analysis.The main results were as follows:1.According to single factor experiment design,the total flavonoids contented in wild chrysanthemums was taken as the research index,and the five factors affecting them were studied.The results showed that the total flavonoids in wild chrysanthemums were extracted best when the ethanol concentration was 40%,the liquid-solid ratio of the concentration of ethanol to the original medicinal material was 50∶1(mL/g),the extraction time on the ultrasound system was 1h,the extraction temperature on the ultrasound system was 35℃and the extraction power of the ultrasonic instrument was 480 W.2.Based on the results of previous tests,the response surface analysis software Design Expert was used to fit and analyze the experiment using Box-Behnken principle.And the regression calculation showed that the total flavonoid extraction effect of wild chrysanthemum flowers was the best when the liquid-solid ratio of 39.89%ethanol to raw herbs was 50.61∶1(mL/g),the ultrasonic extraction time was 59.83 min,the extraction temperature was 35.17℃and the extraction power was 478.06 W.
