血清高三酰甘油对机采血小板保存期间活化损伤的影响研究

摘要 目的:探讨血清高三酰甘油(triglycerides,TG)对机采血小板保存期间活化损伤的影响。方法:选取2021年5—7月来源符合国家标准的血小板捐献者131例的机采血小板,于(22±2)℃震荡保存,根据血清TG水平的不同分为3组,正常组(<1.70 mmoL/L)、临界性高组(1.70~2.25 mmoL/L)和升高组(>2.25 mmoL/L),分别于保存第1、2、3、4、5天进行血小板计数和P选择素(P-selectin,CD62p)、血小板颗粒膜蛋白(platelet granule menbrane protein,CD63)表达水平的测定,对比3组血小板捐献者不同血清TG水平对机采血小板保存期间活性损伤的影响。结果:血清TG临界性高组血小板的CD62p、CD63表达水平相比正常组在保存5 d内无明显变化(P>0.05);血清TG升高组机采血小板的CD62p、CD63表达水平较正常组在保存第2、3、4、5天明显增加(P<0.05)。结论:机采血小板在保存期间的活性受到血小板捐献者血清TG水平的影响,但血清TG处于临界性高水平的血小板捐献者仍能为临床血小板输注提供有力帮助,具有重要临床意义。 Objective:To explore the effect of serum high triglycerides(TG)on the activation and damage of machine-harvested platelets during storage.Methods:Collection in accordance with the national standard of the platelet donors from May to July 2021,131 cases apheresis plateletsand stored at(22±2)℃.The platelet donors were divided into three groups according to different serum TG levels,the normal group(<1.70 mmoL/L)and moderate high group(1.70-2.25 mmoL/L)and the high group(>2.25 mmoL/L).The platelet count and the expression levels of P-selectin(CD62p)and platelet granule menbrane protein(CD63)were measured on the first 1,2,3,4,and 5 days of storage.Analyze the effects of different serum TG levels of three groups of platelet donors on the activity damage of machine-collected platelets during storage.Results:The expression levels of CD62p and CD63 of platelets in the moderate high group did not change significantly within 5 days of storage compared with the normal group(P>0.05);compared with the normal group,the CD62p and CD63 expression levels of the machine-collected platelets in the high group were significantly increased on the second,third,fourth,and fifth day of storage(P<0.05).Conclusion:The activity of machine-collected platelets during storage is affected by the platelet donors'serum TG level,but platelet donors whose serum TG level is at a moderate high level can still provide powerful help for clinical platelet transfusion,which has important clinical significance.
