摘要
目的探究高血压阴虚证大鼠主动脉细胞焦亡的潜在机制。方法10只SPF级WKY大鼠作为对照组,20只SPF级SHR大鼠随机分为高血压组和高血压阴虚证组,每组10只。高血压阴虚证组大鼠给予温燥药物灌胃复制阴虚模型,药物组成有淡附片、肉桂、吴茱萸等比例混合,煎煮浓缩药液含生药1 g/mL,按照1 mL/100 g浓度灌胃30 d。实验结束后,HE染色观察大鼠主动脉组织病理变化;RT-qPCR法及Western Blot法检测主动脉细胞焦亡相关因子NLRP3、IL-1β、Caspase-1及GSDMD mRNA及蛋白表达水平。结果HE染色显示高血压组和高血压阴虚证组血管内皮细胞肿胀,高血压阴虚证组损伤更显著。与对照组相比,高血压组和高血压阴虚证组主动脉组织NLRP3、IL-1β、Caspase-1及GSDMD mRNA和蛋白表达水平显著上升。与高血压组相比,高血压阴虚证组主动脉组织Caspase-1、GSDMD mRNA表达水平显著上升。高血压阴虚证组主动脉组织NLRP3、IL-1β、Caspase-1及GSDMD蛋白表达水平显著上升。结论高血压阴虚证大鼠主动脉细胞焦亡程度更显著,血管内皮损伤更严重。
Objective To explore the potential mechanism of cell pyroptosis in aorta of hypertension rats with Yin deficiency syndrome.Methods Ten SPF WKY rats were used as the control group.Twenty SPF SHR rats were randomly divided into two groups including hypertension rats group and hypertension rats with Yin deficiency syndrome group.There were 10 rats in each group.Hypertension rats with Yin deficiency syndrome group were given warm and dry drugs by gavage to duplicate the Yin deficiency syndrome model.The model drug was composed of Danfupian(Radix Aconiti Lateralis Preparata),Rougui(Cinnamomi Cortex)and Wuzhuyu(Euodiae Fructus),which were mixed in the equal proportion.The drug was decocted and concentrated to 1 g of crude drug per milliliter.The rats was administrated by gavage at the concentration of 1 mL/100 g for 30 days.After experiment,HE staining was used to observe the pathological changes of aorta.The expressions of NLRP3,IL-1β,Caspase-1 and GSDMD mRNA and protein were detected by RT-qPCR and Western Blot.Result HE staining showed that the vascular endothelial cells swelled in the hypertension rats group and hypertension rats with Yin deficiency syndrome group.The damage was more significant in the hypertension rats with Yin deficiency syndrome group.Compared with those of the control group,the expression levels of NLRP3,IL-1β,Caspase-1 and GSDMD mRNA and protein in aorta of hypertension rats group and hypertension rats with Yin deficiency syndrome group were significantly increased.Compared with those of the hypertension rats group,the expression levels of Caspase-1 and GSDMD mRNA in aorta of hypertension rats with Yin deficiency syndrome group were increased significantly.The protein expression levels of NLRP3,IL-1β,Caspase-1 and GSDMD in aorta of hypertension rats with Yin deficiency syndrome group were increased significantly.Conclusion The degree of cell pyroptosis in hypertension rats with Yin deficiency syndrome was more significant,and vascular endothelial injury was more serious.
