不同辅料炮制对吴茱萸指标性成分及体外肝细胞毒性的影响 被引量:3

Study on Index Components and Hepatotoxicity in Vitro in Wuzhuyu(Euodiae Fructus)Based on Different Adjuvants Processing

摘要 目的研究不同辅料炮制对吴茱萸指标性成分含量以及体外肝毒性作用的影响,初步解析炮制原理。方法制备吴茱萸生品以及吴茱萸不同辅料炮制品(甘草制吴茱萸、盐炙吴茱萸、酒炙吴茱萸、醋炙吴茱萸、黄连炙吴茱萸、姜炙吴茱萸),采用《中华人民共和国药典》法测定浸出物含量、挥发油含量,以及柠檬苦素、吴茱萸碱、吴茱萸次碱含量,采用CCK-8法测定L02肝细胞的细胞抑制率(%),并测定细胞上清液中乳酸脱氢酶(LDH)活性。结果与吴茱萸生品比较,不同辅料炮制都能提高浸出物含量、降低挥发油含量,但对柠檬苦素、吴茱萸碱、吴茱萸次碱含量,不同辅料炮制有不同程度的影响;不同辅料炮制后均能降低对L02肝细胞的毒性作用,其中以姜炙吴茱萸、甘草制吴茱萸和盐炙吴茱萸的减毒作用较好,酒炙吴茱萸、醋炙吴茱萸和黄连炙吴茱萸的减毒作用次之。结论不同辅料炮制会对吴茱萸的指标性成分有不同程度的影响,同时也会造成体外肝毒性减毒效果的不同程度变化。 Objective To study the impact of the index components and hepatotoxicity in vitro in Wuzhuyu(Euodiae Fructus)based on different adjuvants processing and preliminarily analyze the processing principle.Method Preparation of Wuzhuyu(Euodiae Fructus)raw product and different adjuvants processed products[Gancao(Glycyrrhiza)-processed product,salt-processed product,wine-processed product,vinegar-processed product,Huanglian(Rhizoma Coptidis)-processed product,Shengjiang(Ginger)juice-processed product].The contents of extracts,volatile oil,limonin,evodiamine and rutaecarpine of Wuzhuyu(Euodiae Fructus)processed products were determined by pharmacopoeia method.The cell inhibition rate(%)of L02 liver cells was determined by CCK-8 method and the activity of lactate dehydrogenase(LDH)in the supernatant was determined.Result Compared with Wuzhuyu(Euodiae Fructus)raw product,the processing of different adjuvants could increase the contents of extracts and reduce the content of volatile oil,but the processing of different adjuvants had different degrees of impact on the contents of limonin,evodiamine and rutaecarpine.The processing of different adjuvants could reduce hepatotoxicity of L02 cells,among which,Shengjiang(Ginger)juice-processed product,Gancao(Glycyrrhiza)-processed product and salt-processed product had the better effect,followed by wine-processed product,vinegar-processed productv and Huanglian(Rhizoma Coptidis)-processed product.Conclusion The processing of different adjuvants have different effects on the chemical composition of Wuzhuyu(Euodiae Fructus),and also caused different degrees of changes in hepatotoxicity in vitro.
