摘要
氯喹(Chloroquine,CQ)是一种经典的自噬抑制剂,通过抑制溶酶体活性来抑制细胞自噬。该文利用多种光谱法,探究氯喹与人血清白蛋白(Human serum albumin,HSA)的相互作用。结果表明,CQ能通过静态猝灭模式有效猝灭HSA的内源荧光。通过获取作用过程中的热力学参数,可得到两者结合的主要作用力为氢键和范德华力,且为自发反应。同时,CQ会造成HSA的色氨酸残基周围亲水性增大,并引起HSA二级结构的变化。
Chloroquine(CQ)is a classical autophagy inhibitor,which blocks autophagy by inhibiting lysosomal acidification.In this paper,the interaction between CQ and human serum albumin(HSA)was studied by various spectroscopies.The results showed that CQ could effectively quench the endogenous fluorescence of HSA by static quenching mode.It can be speculated from the thermodynamic parameters of the reaction process that the interaction between CQ and HSA is spontaneous,and the main binding force is hydrogen bond and van der Waals force.In addition,CQ could increase the hydrophilicity around the tryptophan residue of HSA as well as change the secondary structure of HSA.
