2020年黑河市某规模化牛场BAstV检测及ORF1a基因遗传变异分析 被引量:1

Detection and Genetic Evolution Analysis of the ORF1a Gene of BAstV at a Cattle Farm in the Heihe City in 2020

摘要 为调查2020年黑河市某规模化奶牛养殖场中牛星状病毒(bovine astrovirus,BAstV)的感染及遗传进化情况,应用RT-PCR方法对该牛场的腹泻样品进行检测,并对BAstV ORF1a基因进行扩增、测序以及遗传进化分析。结果表明,15份腹泻粪便样品中,BAstV的检测阳性率为20%(3/15),对测序成功的2株BAstV毒株进行序列分析显示,鉴定的2株BAstV毒株的核苷酸与氨基酸同源性分别为99.3%和99.4%。2株毒株都与我国BAstV四川流行毒株,登录号NC_037655.1核苷酸同源性较高,分别为96.6%和97.3%,而其与嗜神经型BAstV欧洲毒株,KX266908.1差异较大,核苷酸同源性分别为36.7%和37.4%。系统发育进化树结果显示,试验鉴定的2株BAstV与已报道的其他中国BAstV流行参考毒株关系密切。 To investigate the infection and genetic evolution of Bovine astrovirus(BAstV)in a large-scale dairy farm in Heihe City city in 2020,RT-PCR was used to detect diarrhea samples from the cattle farm,and the BAstV ORF1a gene was amplified,sequenced and genetic evolution analyzed.The results showed that the positive rate of BAstV was 20%(3/15)in 15 diarrhea samples.The sequence analysis of the two BAstV strains successfully sequenced showed that the nucleotide and amino acid homology of the two BAstV strains were 99.3%and 99.4%,respectively.The nucleotide homology of the two strains was 96.6%and 97.3%,respe-ctively,with BAstV sichuan Sichuan province epidemic strain(registration number NC_037655.1)in China.However,the nucleotide homology of the two strains was 36.7%and 37.4%,respectively,with BAstV European strain(KX266908.1).Phylogenetic tree results showed that the two BAstV strains identified in this study were closely related to other BAstV epidemic reference strains reported in China.Therefore,it is was necessary to prevent and control BAstV in Heilongjiang regionprovince.
