高温胁迫下叶用莴苣核体系酵母文库构建与鉴定 被引量:1

摘要 【目的】为探明高温胁迫下叶用莴苣发生抽薹现象时核体系内蛋白质间相互作用机制。【方法】以GB-30生菜品种为试验材料,以Gateway技术为手段,构建了高温条件下生菜核体系酵母cDNA文库。【结果】构建的核体系酵母文库已达质量标准,总克隆数1.36×10;CFU/mL,重组率为100%,平均插入片段长度>1000 bp。【结论】该试验条件下成功构建了高温胁迫下生菜核体系酵母文库,该文库质量较高,完整度较好,可用于探明高温条件下的蛋白间相互作用的cDNA文库。 [Objective] In order to explore the mechanism of protein-protein interaction in the nuclear system during bolting of leaf lettuce under high temperature stress. [Methods] The yeast cDNA library of lettuce nuclear system under high temperature was constructed by using ‘GB-30’ lettuce variety as experimental materials and gateway technology. [Results] The constructed nuclear system yeast library has reached the quality standard, and the total number of clones is 1.36 × 10;CFU/mL, the recombination rate was 100%, and the average length of the inserted fragment was >1 000 bp. [Conclusion] The yeast library of lettuce nuclear system under high temperature stress was successfully constructed under this experimental condition. The library has high quality and good integrity, and can be used to explore the cDNA library of protein-protein interaction under high temperature.
