胃癌组织中叉头框蛋白M1的表达与临床病理特征及预后的相关性

Correlation of Forkhead Box Protein M1 Expression with Clinicopathological Features and Prognosis in Gastric Cancer

目的探讨胃癌组织中叉头框蛋白M1(FOXM1)的表达及其与预后的相关性。方法采用免疫组化SP法检测并比较胃癌和癌旁组织FOXM1的表达。分析FOXM1表达与胃癌患者临床病理特征的关系;采用Cox比例风险回归模型分析胃癌患者预后的危险因素;采用Kaplan-Meier生存曲线分析FOXM1与患者预后的关系。结果胃癌组织FOXM1蛋白高表达率高于癌旁组织,差异有统计学意义(P<0.05);胃癌组织FOXM1高表达与年龄、分化程度、TNM分期、淋巴结转移、肿瘤大小、脉管神经侵犯和浸润深度相关(P<0.05);Cox回归分析显示,TNM分期、淋巴结转移、脉管神经侵犯、浸润深度和FOXM1均是影响胃癌患者无进展生存期(PFS)和总体生存时间(OS)的独立危险因素(P<0.05);Kaplan-Meier法分析结果显示,FOXM1高表达组患者PFS和OS累积生存率低于低或无表达组(P<0.05)。结论FOXM1可能在胃癌的发生发展过程中发挥着重要作用,且FOXM1高表达是胃癌的不良预后因素。

Objective To explore the expression of forkhead box M1(FOXM1)in gastric cancer and its correlation with prognosis.Methods Immunohistochemical SP method was used to detect expression of FOXM1 in gastric cancer and adjacent tissues.Relationship between FOXM1 and clinicopathological features of gastric cancer was analyzed.Cox proportional hazard regression model was used to analyze risk factors for prognosis of gastric cancer patients.Kaplan Meier survival curve was used to analyze relationship between FOXM1 and prognosis.Results The high positive expression rate of FOXM1 in gastric cancer group was higher than that in paracancerous tissues(P<0.05).The high positive expression of FOXM1 in gastric cancer was associated with age,differentiation,TNM stage,lymph node metastasis,tumor size,vascular invasion and depth of invasion(P<0.05).Cox regression analysis showed that TNM stage,lymph node metastasis,vascular invasion,depth of invasion and FOXM1 were independent risk factors for progression free survival(PFS)and overall survival(OS)of gastric cancer patients(P<0.05).Kaplan-Meier analysis showed that cumulative survival rates of PFS and OS in FOXM1 overexpression group were lower than that in low or no expression group(P<0.05).Conclusion FOXM1 protein may be highly expressed in gastric cancer tissue which could be closely related to occurrence,development and prognosis of gastric cancer.
