摘要
目的:验证肺炎支原体(MP)感染人肺微血管内皮细胞(HPMEC)及小鼠后上调炎症因子水平以及清肺通络方对炎症因子的下调作用。方法:实验分为正常对照组、模型组、清肺通络方组。用MP菌株感染模型组及药物小鼠及HPMEC细胞后,用清肺通络方干预药物组小鼠及HPMEC细胞。以ELISA法检测HPMEC细胞上清液及各组小鼠血清中TGF-β、IL-1β水平。结果:与正常对照组比较,模型组细胞上清液及小鼠血清中TGF-β、IL-1β水平均显著升高(P<0.01)。与模型组比较,清肺通络方组血清中TGF-β、IL-1β水平均显著降低(P<0.01)。结论:MP可引起细胞上清液及小鼠血清中炎症因子TGF-β、IL-1β表达上调,清肺通络方具有减轻炎症水平的作用。
Objective: To establish a mycoplasma pneumoniae(MP)-induced apoptosis model of human pulmonary microvascular endothelial cells(HPMEC) and a mouse model of MP infection, to verify that MP infects HPMEC cells and up-regulates the levels of inflammatory factors in mice, and Qingfei Tongluo Formula down-regulation of inflammatory factors.Methods: The mice were divided into normal control group, model group, and drug group. After infection of the model group,drug mice and HPMEC cells, Qingfei Tongluo Formula was used to intervene drug group mice and HPMEC cells. ELISA method was used to detect the levels of TGF-β and IL-1β in mouse serum and HPMEC cell supernatant of each group. Results:Compared with the control group, the levels of TGF-β and IL-1β in the cell supernatant and serum of the model group were higher than those in the control group(P<0.01). Compared with the model group, the levels of TGF-β and IL-1β in the serum were significantly reduced(P<0.01). Conclusion: MP can increase the expression of inflammatory factors TGF-β and IL-1β in the cell supernatant and mouse serum. Qingfei Tongluo Formula has the effect of reducing the level of inflammation.
