Multi-omics analysis of yin deficiency and internal heat syndrome of non-small cell lung cancer based on system biology

摘要 目的:探讨阴虚内热型非小细胞肺癌肠道菌群、代谢组学和蛋白组学的基本特征。方法:选择阴虚内热型非小细胞肺癌患者15例作为阴虚内热证肺癌组,选择健康志愿者15名作为健康对照组,收集两组粪便标本,用16S rDNA技术检测肠道微生态。采集两组血清标本,通过液相色谱-质谱联用技术比较两组代谢组学和蛋白质组学差异。最后通过WGCNA软件对多组学结果进行联合分析。结果:两组间存在6种差异菌群,9种差异蛋白(上调5个,下调4个),9种差异代谢物(负离子模式下3个,正离子模式下6个),差异菌群、差异蛋白与差异代谢物间存在相关性,联合分析发现差异物质与癌症通路相关。结论:阴虚内热证非小细胞肺癌具有特异的肠道菌群、蛋白质和代谢物,与肿瘤的发生发展密切相关。 Objective: To explore the basic characteristics of intestinal flora, metabolomics and proteomics of non-small cell lung cancer with yin deficiency and internal heat syndrome. Methods: A total of 15 patients with yin deficiency and internal heat syndrome of non-small cell lung cancer were selected as the yin deficiency and internal heat syndrome lung cancer group, and 15 healthy volunteers were selected as the healthy control group. Fecal samples of the two groups were collected and intestinal microecology was detected by 16s rDNA technique. Serum samples were collected from the two groups, and the differences in metabolomics and proteomics were compared by liquid chromatography-mass spectrometry. Finally, the joint analysis of the results of multiple omics was conducted through WGCNA software. Results: There were 6 different microflora between the two groups, 9 differential proteins(5 up-regulated and 4 down-regulated), 9 differential metabolites(3 in negative ion mode and 6 in positive ion mode). There was a correlation between differential bacteria, differential proteins and differential metabolites.Joint analysis found that differential substances were related to cancer pathway. Conclusion: Non-small cell lung cancer with yin deficiency and internal heat syndrome has specific flora, proteins and metabolites, which is closely related to the occurrence and development of the tumor.
