益气养阴消瘿方对自身免疫性甲状腺炎模型大鼠脾脏Breg细胞的调节作用

Regulating effect of Yiqi Yangyin Xiaoying Formula on spleen Breg cell of autoimmune thyroiditis model rats

目的:研究益气养阴消瘿方对自身免疫性甲状腺炎(AIT)模型大鼠脾脏Breg细胞的调节效应。方法:雌性SD大鼠随机分为正常对照组,模型组,益气养阴消瘿方低、中、高剂量组,以皮下注射猪甲状腺球蛋白,并配合高碘水喂养制造AIT模型。ELISA法检测各组大鼠血清促甲状腺激素(TSH)、甲状腺球蛋白抗体(TGAb)及甲状腺过氧化物酶抗体(TPOAb)水平,流式细胞术检测大鼠脾脏B细胞和调节性B细胞(Breg)比例。结果:与正常对照组比较,模型组大鼠血清TSH、TGAb、TPOAb水平显著升高(P<0.01),而脾脏B细胞、Breg细胞比例及Breg/B比值显著降低(P<0.05);与模型组比较,益气养阴消瘿方各剂量组大鼠血清TSH、TGAb、TPOAb水平显著下降(P<0.01),且脾脏B细胞、Breg细胞比例及Breg/B比值均显著上调(P<0.05)。结论:益气养阴消瘿方可降低AIT大鼠TSH、TGAb、TPOAb水平,改善免疫失衡状态,可能与其调节脾脏Breg细胞表达有关。

Objective: To investigate the regulating effect of Yiqi Yangyin Xiaoying Formula on spleen Breg cell in autoimmune thyroiditis(AIT) model rats. Methods: Female SD rats were randomly divided into normal control group, model group, Yiqi Yangyin Xiaoying Formula groups(low, medium and high dose). The AIT model was made by subcutaneous injection of porcine thyroglobulin and feed with high iodine water. The levels of serum TSH, TGAb and TPOAb were detected by ELISA.The ratios of B cell and Breg cell in spleen were detected by flow cytometry. Results: The concentration of serum TSH, TGAb and TPOAb in the model group were significantly higher than those in control group(P<0.01), while the level of spleen B cell, Breg and Breg/B ratio were decreased in the model group than those in control group(P<0.05). Compared with the model group, the concentration of serum TSH, TGAb and TPOAb were lowered(P<0.01), while the level of spleen B cell, Breg and Breg/B ratio were upregulated in the Yiqi Yangyin Xiaoying Formula groups(P<0.05). Conclusion: Yiqi Yangyin Xiaoying Formula could reduce the levels of serum TSH, TGAb and TPOAb in AIT rats and attenuate immune imbalance, which may be related to the regulation of spleen Breg expression.
