摘要
为探讨FoxO1基因多态性与藏鸡生长和屠宰性状之间的关系,试验以藏鸡为研究对象,采用限制性片段聚合酶链式反应(PCR-RFLP)技术检测藏鸡FoxO1基因的遗传多态性。结果显示:在FoxO1基因第一内含子上检测到5个多态性位点;在3187T>G位点,GT基因型个体的体斜长、龙骨长和胸深均显著高于TT基因型个体(P<0.01或P<0.05),TT基因型个体的活体重、腿肌重、胸肌重、全净膛重均极显著低于GT基因型(P<0.01),半净膛重显著低于GG和GT基因型(P<0.05);在40088A>G位点,AA基因型个体的活体重显著高于GG基因型个体(P<0.05);在49628C>T位点,CC基因型个体的胸宽显著高于TC基因型个体(P<0.05)和TT基因型个体(P<0.01);在50344C>T位点,TC基因型个体的腿肌重显著高于CC基因型个体(P<0.05);在52564G>T位点,GG和TG基因型个体的屠宰率显著高于TT基因型个体(P<0.01)。连锁不平衡分析表明,5个SNPs位点间连锁程度弱。研究提示,5个SNPs中有4个SNPs位点至少与一个生长或屠宰性状有关联,FoxO1基因有望作为个体经济性状的有效候选基因进行深入研究。
This experiment took Tibetan chicken as the materials, PCR-RFLP was used to detect the SNP of FoxO1 gene, the correlation of between the SNP genotypes of FoxO1 gene with growth and slaughter traits was analyzed. The results showed that the 5 SNPs mutation sites were detected in intron 1. At the 3187T>G position, the body oblique length, keel length and chest depth of individuals with GT genotype were significantly higher than those individuals with TT genotype(P<0.01 or P<0.05);The live weight, carcass weight, leg muscle weight, breast muscle weight, full eviscerated weight of individuals with TT genotype were very significantly lower than those individuals with GT genotype(P<0.01) and the half-eviscerated weight of individuals with TT genotype significantly lower than those individuals with GG and GT genotype(P<0.05);At the 40088A>G position, the live weight of individuals with AA genotype were significantly higher than those individuals with GG genotypes(P<0.05);At the 49628C>T position, the chest width of individuals with CC genotype was significantly higher than those individuals with TC genotype(P<0.05) and individuals with TT genotype(P<0.01);At the 50344C>T position, the leg muscle weight of individuals with TC genotype were significantly higher than individuals with CC genotype(P<0.05);At the 52564G>T position, the slaughter rate of individuals with GG and TG genotypes was significantly higher than individuals with TT genotype(P<0.01). Linkage disequilibrium analysis showed that the 5 SNPs did not reach the condition of linkage disequilibrium, the degree of linkage was weak. The results suggested that among the 5 SNPs, 4 SNPs were associated with at least one growth or slaughter trait of Tibetan chicken, and FoxO1 gene was expected to be an effective candidate gene for individual economic traits for in-depth study.
