免疫INHα亚基对浙东白鹅产蛋性能和等级卵泡发育的影响

Effects of Active Immunization Against INHα Subunit on Egg Laying and Hierarchical Follicle Development in Zhedong White Goose

摘要 为了揭示抑制素对鹅产蛋性能和等级卵泡发育的影响,研究选取60只同批次即将开产的浙东白鹅(165日龄),平均分为2组,每组30只,试验组每只鹅免疫INHα亚基重组蛋白1 mg,对照组每只鹅免疫牛血清白蛋白1 mg。首先观察了第一个产蛋-就巢周期内产蛋和就巢情况以及血浆孕酮浓度变化,其次在第二个产蛋-就巢周期内检测了卵巢不同等级卵泡数量和等级卵泡颗粒层孕酮合成(FSHR、LHR、StAR、CYP11A1和3β-HSD)、卵黄沉积(OCLN)和细胞凋亡(FoxO3、Bax、Bcl-2和Caspase3)等相关基因的表达量变化。结果显示:免疫INHα亚基降低浙东白鹅的产蛋量和血浆孕酮水平,延迟就巢发生,增加等级前卵泡数量,下调等级卵泡颗粒层中LHR mRNA表达量,上调OCLN和FoxO3 mRNA表达量。结果表明INHα亚基在促进鹅等级卵泡颗粒层孕酮合成、卵黄沉积和细胞增殖方面发挥着重要作用,也表明免疫INHα亚基不利于家禽卵巢等级卵泡发育。 In order to reveal the effects of inhibin on egg laying and hierarchical follicle development in geese, sixty 165-day-age pre-laying Zhedong White geese with the same genetic background were selected and equally divided into two groups with 30 geese in each group. The treatment group was actively immunized against the recombinant goose INHα subunit protein(1mg per goose), whereas the control group was immunized against bovine serum albumin(1 mg per goose). This study observed egg laying, incubation, and plasma progesterone concentration of geese during the first egg-laying cycle. The number of ovarian follicles and the mRNA expression level of genes related to progesterone synthesis(FSHR, LHR, StAR, CYP11A1, and 3β-HSD), yolk deposition(OCLN) and apoptosis(FoxO3, Bax, Bcl-2, and Caspase3) in the granulosa layer of ovarian hierarchical follicles were analyzed during the second egg-laying cycle. The results showed that active immunization against INHα subunit decreased egg production and plasma progesterone concentration, postponed incubation behavior, increased the number of pre-hierarchical follicles, and down-regulated the mRNA expression level of LHR gene and up-regulated mRNA expression levels of OCLN and FoxO3 genes in the granulosa layer of hierarchical follicles in geese. These results suggest that INHα subunit played an important role in promoting progesterone synthesis, yolk deposition and cell proliferation in the granulosa layer of hierarchical follicles in geese. The study was for the first time to reveal the adverse effects of active immunization against INHα subunit on the development ovarian hierarchical follicles in poultry.
