不同品规鹿茸的抗氧化活性及其对RAW264.7细胞的免疫调节作用考察 被引量:1

摘要 目的研究不同品规鹿茸的抗氧化活性以及对RAW264.7细胞的免疫调节作用的差异,为不同品规鹿茸的差异分析提供数据支持。方法测定了不同品规鹿茸的抗1,1-二苯基-2-三硝基苯肼(DPPH)、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)、羟基(OH)自由基活性,噻唑蓝(MTT)比色法测定不同品规鹿茸对RAW264.7细胞增殖能力的影响、中性红吞噬实验测定不同品规鹿茸对RAW264.7细胞吞噬能力影响并进一步测定不同品规鹿茸对RAW264.7细胞白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)mRNA表达量的影响。结果不同品规鹿茸抗氧化能力各有差异。促细胞IL-1β、IL-6、TNF-αmRNA表达能力结果均显示出二杠茸高于毛桃茸、三岔茸,头茬茸高于二茬茸的趋势。结论不同品规鹿茸抗氧化活性各有差异,二杠茸、头茬茸对RAW264.7细胞的免疫调节作用高于毛桃茸、三岔茸和二茬茸。 Objective To study the the difference of anti-oxidant activity,immunomodulatory effect on RAW264.7 cells and provide data support for the analysis of velvet antler of different grades.Methods The anti-1,1-diphenyl-2-picrylhydrazyl(DPPH),2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS)and hydroxyl(OH)free radical activities of deer antlers of different grades were tested.Methylthiazolyldiphenyl-tetrazolium bromide(MTT)method was used to determine the effect on the proliferation of RAW264.7 cells.The neutral red phagocytic experiment was used to determine the effect on the phagocytic ability of RAW264.7 cells.The effect of velvet antler of different grades on the expression of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)mRNA in RAW264.7 cells was measured.Results The antioxidant capacity of antler of different grades was different.The results of the expression of IL-1β,IL-6,and TNF-αmR-NA in the cells showed that the two-branched velvet antler was higher than one-branched velvet antler and three-branched velvet antler and the first-cut velvet antler was higher than the reborn velvet antler.Conclusion The antioxidant activity of dif-ferent grades of velvet antler is different and the immunomodulatory effect of two-branched velvet antler and first-cut velvet antler on RAW264.7 cells is higher than that of one-branched velvet antler,three-branched velvet antler and reborn velvet antler.
