摘要
目的探讨DJ-1表达下调对人支气管上皮细胞(HBE细胞)生物学行为的影响。方法构建DJ-1基因siRNA慢病毒载体,感染HBE细胞(DJ-1 siRNA组),并设置Control siRNA慢病毒载体对照(Control siRNA组)及空白对照(BC组),分别采用流式细胞术检测各周期细胞,四甲基偶氮唑盐(MTT)比色法检测细胞增殖情况,克隆形成试验检测细胞克隆情况,Transwell小室试验检测细胞迁移情况。结果DJ-1 siRNA组G_(1)期细胞比例高于BC组、Control siRNA组,S期、G_(2)期细胞比例低于BC组、Control siRNA组,差异有统计学意义(P<0.05)。与Control siRNA组和BC组比较,DJ-1 siRNA组在接种后24、48、72、96 h的吸光度值明显降低,差异有统计学意义(P<0.05);DJ-1 siRNA组在接种后48、72、96 h的增殖抑制率分别是36.9%、60.8%和37.6%。DJ-1 siRNA组的克隆数、迁移细胞数明显少于Control siRNA组与BC组,差异有统计学意义(P<0.05)。结论下调DJ-1的表达具有抑制HBE细胞分裂、增殖、迁移和侵袭的作用。
Objective To investigate the effect of down-regulation of DJ-1 expression on the biological behavior of human bronchial epithelial cells(HBE cells).Methods Constructed DJ-1 gene siRNA lentiviral vector,infected HBE cells(DJ-1 siRNA group),and set Control siRNA lentiviral vector control(Control siRNA group)and blank control(BC group).Flow cytometry was used to detect cells in each cycle,MTT colorimetry method used to detect cell proliferation,clone formation assay was used to detect cell clones,and Transwell chamber assay was used to detect cell migration.Results The proportion of cells in G_(1) phase in DJ-1 siRNA group was higher than that in BC group and Control siRNA group,and the proportion of cells in S phase and G_(2) phase was lower than that in BC group and Control siRNA group,and the differences were statistically significant(P<0.05).Compared with the Control siRNA group and the BC group,the absorbance values of the DJ-1 siRNA group at 24,48,72 h and 96 h after vaccination were significantly decreased,and the differences were statistically significant(P<0.05).The proliferation inhibition rates of the DJ-1 siRNA group at 48,72 h and 96 h after vaccination were 36.9%,60.8% and 37.6% respectively.The number of clones and migrated cells in the DJ-1 siRNA group were significantly less than those in the Control siRNA group and the BC group,and the differences were statistically significant(P<0.05).Conclusion Down-regulation of DJ-1 expression inhibited the division,proliferation,migration and invasion of HBE cells.
