摘要
目的:通过16S rDNA技术,进行高通量测序分析,从微观角度探讨不同性别痰湿体质受试者肠道菌群的特征和差异。方法:60位20~50岁痰湿体质受试者,男女各半。严格按照纳入标准和排除标准收集60份粪便标本。Illumina MiSeq高通量测序平台被用于在338F_806R区域进行16S rDNA测序,以检测肠道菌群。结果:偏最小二乘回归分析法(PLS-DA)分析显示,男女受试者明显区分并聚成两个类群,可以明显区分为两个不同的组,说明痰湿体质男性和女性肠道菌群组成在OTU水平上具有显著差异。菌群Alpha多样性分析显示,女性痰湿体质者肠道菌群多样性高于男性。肠道菌群的群落组成分析结果显示,在门水平上,经Kruskal-Wallis秩和检验,梭菌门、Tenericutes门、蓝细菌门(0.01<P≤0.05)为男女受试者肠道菌群在门水平差异明显的菌门。拟杆菌属(0.01<P≤0.05)、大肠杆菌-志贺菌属(0.001<P≤0.01)为男女受试者肠道菌群在属水平差异明显的菌属。结论:男女痰湿体质受试者肠道菌群的不同可能是代谢性疾病及其合并症的发生率存在性别差异的原因。
Objective:To explore the features and differences of intestinal flora in people with phlegm-dampness constitution of different genders in a microcosmic perspective,using 16S rDNA technology to perform high-throughput sequencing analysis. Methods:Sixty subjects of phlegm-dampness constitution aged 20-50 years were enrolled,30 males and 30 females.Sixty stool specimens were collected strictly in accordance with inclusion criteria and exclusion criteria. Illumina MiSeq high-throughput sequencing platform was applied to process 16S rDNA sequencing in Area 338F_806R for intestinal flora detection. Results:PLS-DA analysis showed that male and female subjects were clearly distinguished and differentiated into two groups,which could be clearly distinguished into two different groups. This showed that the compositions of intestinal flora were significant different at OTU levels in the male and female subjects with phlegm-dampness constitution. According to the analysis of bacterial flora Alpha the diversity of intestinal flora in the female was more than that in the male. The composition analysis of intestinal flora revealed that at the phylum level,clostridium,tenericutes,and cyanobacteria(0.01<P≤0.05) were the intestinal flora with significant difference in male and female subjects,and at the genus level,bacteroides(0.01<P≤0.05),escherichia-shigella(0.001<P≤0.01),were the intestinal flora with obvious differences in the male and the female. Conclusions:The difference in intestinal flora of male and female subjects with phlegm-dampness constitution may be the cause of gender differences in the incidence of metabolic diseases and their complications.
