摘要 依托教师的科研成果,设计了“超顺磁性纳米簇捕获和分离细菌综合性实验”。利用高温水解法制得超顺磁性纳米簇,进一步修饰沙门氏菌抗体,构建磁性纳米探针,表征了其形貌、磁性和生物活性,并详细探究了对沙门氏菌的分离效率、特异性及抗干扰能力。实验结果表明:磁性纳米探针能够快速、特异、高效地捕获和分离沙门氏菌,捕获效率达95%以上,可以直接用于牛奶、血清等复杂体系。该实验内容完整,具有较高的交叉性和前沿性,有利于培养学生的综合实验能力和科研素养,适合在材料、化学、生物、医学等相关专业高年级本科生中开展。 Based on teachers’scientific research achievements,a comprehensive experiment of capture and separation of bacteria with superparamagnetic nanoclusters is developed.The superparamagnetic nanoclusters are prepared with high temperature hydrolysis method,and by further modification with anti-Salmonella antibody,magnetic nanoprobes are constructed.Their morphology,magnetic property,and biological activity are characterized.The capture efficiency to Salmonella,specificity and anti-interference ability of the magnetic nanoprobes are investigated in detail.Experimental results show that the magnetic nanoprobes can quickly,specifically,and efficiently capture and separate Salmonella with efficiency of more than 95%,and can be directly used in complex matrix such as milk and serum.This experiment has abundant content with highly multi-field crossing and prior.It is beneficial to cultivate students’comprehensive experimental ability and scientific literacy,which is suitable for senior undergraduates majoring in material science,chemistry,biology,medicine and so on.
