脂多糖刺激对断奶仔猪空肠形态结构、炎症反应和程序性坏死信号通路相关基因表达的影响 被引量:6

Effects of Lipopolysaccharide Challenge on Jejunal Morphology,Inflammatory Response and Necroptosis Signaling Pathway Related Gene Expression of Weaned Piglets

摘要 本试验旨在研究脂多糖(LPS)刺激对断奶仔猪空肠形态结构、炎症反应和程序性坏死信号通路相关基因表达的影响。选取12头平均体重为(7.07±0.57)kg的28日龄杜×长×大三元断奶仔猪,随机分为2组,分别为对照组和LPS刺激组,每组6头。预饲9 d后,LPS组腹腔注射100μg/kg BW的LPS,对照组注射等量的生理盐水。LPS注射4 h后将所有仔猪麻醉屠宰,取空肠样品待测。结果表明:与对照组相比,LPS刺激后仔猪空肠绒毛严重萎缩,并大量脱落,绒毛高度和隐窝深度显著降低(P<0.05);空肠黏膜RNA/DNA显著下降(P<0.05),蛋白质含量和蛋白质/DNA有显著下降趋势(P=0.054、P=0.095);空肠炎性细胞因子环氧合酶2(COX2)、热休克蛋白70(HSP70)和白细胞介素-6(IL-6)的mRNA相对表达量显著或极显著升高(P<0.05或P<0.01);程序性坏死信号通路相关基因受体相互作用蛋白激酶1(RIP1)和受体相互作用蛋白激酶3(RIP3)的mRNA相对表达量显著或极显著升高(P<0.05或P<0.01)。由此可见,LPS刺激造成了断奶仔猪空肠损伤和炎症,同时激活了程序性坏死信号通路。 The aim of this study was to investigate the effects of lipopolysaccharide(LPS)challenge on jejunal morphology,inflammatory response and necroptosis signaling pathway related gene expression of weaned piglets.Twenty weaned piglets with the average body of(7.1±0.9)kg and aged 28 days were randomly divided into 2 groups including control group and LPS challenge group,and each group had 6 piglets.Afer 9 day of preliminary feeding,the piglets in the LPS challenge group were treated with intraperitoneal injection of100μg/kg BW LPS,and the those in the control group were injected with the same amount of saline.Piglets were anesthetized for slaughter after 4 h post-injection,and the jejunum sample was taken for further test.The results showed as follows:compared with the control group,LPS challenge caused the jejunal villous atrophy and shedding,and significantly decreased the villous height and the crypt depth of piglets(P<0.05);LPS challenge significantly reduced the jejunal mucosa RNA/DNA(P<0.05),and had significant decreasing trend on protein and protein/DNA(P=0.054,P=0.095);LPS also significantly increased the mRNA relative expression levels of jejunal inflammatory factors including cyclooxygenase 2(COX2),heat shock protein 70(HSP70)and interleukin-6(IL-6)(P<0.05);moreover,LPS significantly increased the mRNA relative expression levels of necroptosis signaling pathway related genes receptor including receptor-interacting protein kinase 1(RIP1)and receptor-interacting protein kinase 3(RIP3)(P<0.05).In conclusion,LPS challenge causes jejunal injury and inflammation,and activates the necroptosis signaling pathway of weaned piglets.
