摘要 建立云南地方品种生姜快繁体系,为姜种的快速繁殖提供技术保障,同时为生产应用提供技术支撑。以云南文山地方品种小黄姜为实验材料,研究其组培快繁技术。结果表明:最佳消毒方式是75%酒精消毒20 s+2%次氯酸钠消毒15 min+无菌水清洗3次;最佳诱导培养基是MS+6-BA 1.0 mg/L+NAA 0.1 mg/L+琼脂7 g/L+蔗糖30 g/L,pH 5.8;最佳继代培养基MS+琼脂7 g/L+蔗糖30 g/L,扩繁系数达5;最佳生根培养基1/2 MS+NAA 0.1 mg/L+卡拉胶7 g/L+蔗糖30 g/L;最佳移栽基质是营养土∶椰糠1∶1(m∶m),移栽成活率达95%以上。 In order to establish a rapid propagation system for ginger in Yunnan,provide technical support for the rapid propagation production and application of ginger varieties,Yunnan Wenshan local variety ginger were used as the experimental material,the tissue culture and rapid propagation technology of ginger was studied.The results showed that the best disin-fection method was 75%alcohol disinfection for 20 s+2%sodium hypochlorite disinfection for 15min+sterile water cleaning for 3 times;The best induction medium was MS+6-BA 1.0 mg×L^(‒1)+NAA 0.1 mg×L^(‒1)+agar 7 g×L^(‒1)+sucrose 30 g×L^(‒1),coordina-tion:5.8;The best subculture medium was MS+agar 7 g×L^(‒1)+sucrose 30 g×L^(‒1),the propagation coefficient reaches 5;The best rooting medium was 1/2MS+NAA 0.1 mg×L^(‒1)+carrageenan 7 g×L^(‒1)+sucrose 20 g×L^(‒1);The best transplanting substrate was nutrient soil:coconut peat(1∶1),the survival rate of transplanting was more than 95%.
