摘要
为研究儋州鸡鸡冠形态差异和发育与BMP2、CHADL基因的蛋白水平表达关系及变化规律,探索其对儋州鸡鸡冠生长发育的调控作用机制。通过免疫印迹(Western Blot)试验方法,对儋州鸡不同冠型(单冠和玫瑰冠)、不同发育阶段(2、4、5、6月龄)、不同性别(公鸡、母鸡)的鸡冠组织进行BMP2、CHADL基因蛋白水平的表达分析(每组各取5只儋州鸡鸡冠组织重复样本)。结果表明:儋州鸡公鸡的单冠和玫瑰冠组织中BMP2蛋白表达在2、4、6月龄的表达趋势均为由高到低再到高,玫瑰冠组织中差异显著;而CHADL蛋白表达呈上升趋势,但差异不显著;2月龄公鸡单冠组中BMP2蛋白表达水平低于玫瑰冠,4、6月龄则高于玫瑰冠,差异均不显著;2、6月龄公鸡单冠组织中CHADL蛋白表达水平低于玫瑰冠,而4月龄高于玫瑰冠,差异均不显著。性成熟中期(5月龄)儋州鸡公鸡和母鸡单冠组织中BMP2、CHADL蛋白表达水平均高于其在玫瑰冠组中的表达,且BMP2蛋白在公鸡2冠型组织间的表达差异显著,2种蛋白在母鸡2冠型组织间差异均不显著;性成熟中期相同冠型鸡冠组织中公鸡的BMP2蛋白表达水平均低于母鸡,且玫瑰冠中差异显著;CHADL蛋白表达水平在性成熟中期不同性别和不同冠型间表达差异不显著。儋州鸡鸡冠形态的差异和发育阶段的不同与BMP2、CHADL基因的蛋白水平表达有一定的关联性,其中BMP2较为显著。BMP2基因的表达量与儋州鸡鸡冠冠型大小和生长月龄呈负相关,可能在调控鸡冠发育上存在抑制作用,其调控作用在性成熟期间较为显著,并且主要体现在公鸡群体中。
In order to study the relevance of the BMP2 and CHADL protein expression in combs with different morphology and development of Danzhou chicken,explore its regulation mechanism on the growth and development of Danzhou chicken’s comb.Western Blot was used in this study to analyze the protein expression of BMP2 and CHADL in comb with different morphology(single comb and rose comb),different growth stage(2,4,5 and 6 months)and the different gender(male and fe-male)of Danzhou chicken(the number of repeated samples in each group was 5).The result showed that the protein expression level of BMP2 trend at 2,4 and 6 months old was high to low and then to high in single comb and rose comb tissue of male Danzhou chickens,and were significant different in rose comb.The protein expression of CHADL was increasing,but the dif-ference was not significant.The protein expression of BMP2 in single comb tissue was lower than rose comb tissue of 2 months old male Danzhou chickens,and were higher than rose comb tissue of 4 and 6 months old male Danzhou chickens,and the difference were not significant.The CHADL protein expression in single comb tissue was lower than rose comb tissue of 2 and 6 months old male Danzhou chickens,but the expression in single comb tissue was higher than rose comb tissue of 4 months old,difference were not significant.In the middle stage of sexual maturity(5 months old),the protein expression level of BMP2 and CHADL in single comb tissue was higher than rose comb tissue Danzhou chickens,and there were significant difference in male different morphology comb of BMP2,but no significant difference in female.In the middle stage of sexual maturity,the BMP2 protein expression in male Danzhou chicken’s comb tissue was lower than female Danzhou chicken with the same morphological comb,and there were significant different in rose comb,,but no significant difference of CHADL.The different morphology and development of Danzhou chicken’s comb were correlated with the protein expression level of BMP2 and CHADL,and BMP2 was more obvious.The BMP2 protein expression was negatively correlated with morphology size and development of Danzhou chicken’s comb,so BMP2 maybe played a role in inhibiting the morphology and development of combs,its regulation role was more significant during sexual maturity,and it’s mostly in male Danzhou chickens.
