响应面法优化超声辅助法提取青楷槭叶红景天苷工艺研究

摘要 以青楷槭叶为原料,采用超声辅助法提取青楷槭叶中的红景天苷。在单因素实验基础上,应用中心组合实验设计原理,以红景天苷得率为响应值,分别考察了影响提取率的乙醇体积分数、液料比、超声提取时间、超声功率及超声温度5个因素,再采用三因素三水平对超声的三个影响因素(温度、功率、时间)进行响应面优化。结果确定提取过程优化的工艺条件:乙醇体积分数50%、液料比20∶1[V(mL)∶m(g)]、超声提取时间20 min以及超声功率180 W。超声提取法可快速地提取青楷槭叶中的红景天苷,具有省时、高效、稳定等优点。 Salidroside was extracted from the leaves of Acer tegmentosum Maxim.by ultrasonic assisted method.Based on the single-factor experiment,taking extraction yield of salidroside as response value,,the factors affecting the extraction rate ethanol volume fraction,liquid material ratio,ultrasonic extraction time and ultrasonic power ultrasonic temperature were investigated by Box-Benhnken central combination experimental design.Then three factors and three levels were used to op-timize the response surface with three influencing factors of ultrasound(temperature,power and time).The extraction pa-rameters optimized were 50%of ethanol volume fraction,20:1[V(mL)∶m(g)]of liquid material ratio,180 W of ultrasonic irradiation power and 20 min of ultrasonic irradiation time.As a conclusion,it was found that the ultrasonic-assisted extraction was a stable,efficient,and energy-saving technology,which could be used in the extraction of salidroside from the leaves of Acer tegmentosum Maxim.
