构建高效准确小鼠动脉血压测量方法的研究

摘要 目的:探讨构建测定小鼠动脉血压的方法,并验证其准确性和有效性。方法:低浓度七氟醚维持小鼠浅麻醉状态(5%诱导,2%维持)下采用智能无创血压计测量小鼠尾动脉压,构建七氟醚-尾套法,并将测量指标,包括收缩压(SBP)、舒张压(DBP)、平均动脉压(MBP)和心率(HR),与清醒束缚法和股动脉插管法比较。结果:七氟醚-尾套法与清醒束缚法相比,测量达到稳定时间明显缩短(P<0.0001),血压及心率无明显差异(P>0.05),Bland-Altman分析结果显示良好的一致性;与股动脉插管法相比,七氟醚-尾套法所测得小鼠SBP无明显差异统计学意义,而DBP和MBP差异显著(P<0.01),Pearson相关分析表明两种方法测得SBP呈强正相关(r=0.8645,P=0.0056)。结论:七氟醚-尾套法可高效准确地用于小鼠血流动力学参数研究。 Objective:To explorean method to measure the arterial blood pressure of mice to verify its accuracy and effectiveness. Methodes:Under low-concentration sevoflurane-induced mouse light anesthesia(5% induction,2% maintenance),the mouse tail artery pressure was measured with a smart non-invasive sphygmomanometer,the sevofluranetail cuff method was constructed,and its measurement indicators,including systolic blood pressure(SBP),diastolic blood pressure(DBP),mean arterial pressure(MBP)and heart rate(HR)were compared with those measured by awake restraint method and femoral artery catheterization. Results:Compared with the awake-restraint method,the sevoflurane-tail cuff method significantly shortened the time to reach stabilization measurement(P<0.0001),and the blood pressure and heart rate were not statistically significant(P>0.05). The Bland-Altman analysis results showed good agreement. Compared with Femoral artery catheterization method,there was no significantly difference in the results of SBP measured by the sevoflurane-tail cuff method,while the difference between DBP and MBP was significant(P<0.01).Pearson correlation analysis revealed that the SBP measured by the two methods was strongly positive correlation(r=0.8645,P=0.0056). Conclusion:The sevoflurane-tail cuff method constructed in this article can be used efficiently and accurately for the study of mice hemodynamic parameter.
