二乙基亚硝胺诱导斑马鱼肝癌模型的探索与优化 被引量:1

摘要 斑马鱼作为一种模式生物,具有其他动物模型所不具备的高通量等优势,非常适用于疾病模型的构建与药物筛选。采用斑马鱼构建肝癌疾病研究平台:以二乙基亚硝胺(DEN)作为癌症诱导剂按低(50μg/mL)、中(100μg/mL)、高(150μg/mL)3个浓度梯度对斑马鱼进行给药处理,诱导斑马鱼肝脏癌症的发生;同时采用石蜡切片、HE染色的方式观察肝脏细胞的癌变过程,并对其他相应的评价指标进行量化处理。结果表明,DEN能显著诱导斑马鱼肝脏发生脂肪组织病变、腹腔积液、肝脏癌变等病理过程,该模型与现有的模型相比,评价方法更加快速、经济,评价指标更完善。 Zebrafish,as a model organism,has the advantages of high throughput compared with the other animals used in the in vivo test.It is very suitable for the establishment of disease models and drug screening.In this study,zebrafish is used to construct a research platform for HCC:diethyl nitrosamine(DEN)is used to induce liver cancer in zebrafish with three concentrations:low(50μg/mL),medium(100μg/mL)and high(150μg/mL);meanwhile,paraffin sectioning and hematoxylin-eosin(HE)staining are performed to observe the canceration of liver cells.Furthermore,the HCC is evaluated and quantified with optimized parameters.The results show that DEN can significantly induce the pathological changes of zebrafish liver,including adipose tissue lesions,ascites,and liver carcinogenesis.Compared with the other existing evaluation method,our zebrafish model is faster and more economical with more quantitative parameters optimized.
