复方透骨草酊对睾酮的抑制及促进毛发生长作用的初步研究

Investigation on inhibiting testosterone and promoting hair growth by compound Tougucao tincture:a preliminary study

目的 探究复方透骨草酊对睾酮诱导的C57BL/6病理脱发模型小鼠的性激素水平、毛发生长及毛囊细胞凋亡情况的作用。方法 将40只小鼠随机分为空白对照组、模型对照组、米诺地尔酊组、复方透骨草酊组,分别在脱毛区外给药,摘眼球取血法检测小鼠血清睾酮、雌二醇、双氢睾酮水平,原位杂交技术原位末端标记法(TUNEL)评价毛囊细胞凋亡水平。结果 与模型对照组相比,复方透骨草酊在毛发生长时间、血清睾酮及双氢睾酮水平、细胞凋亡指数方面均有明显统计学差异(P <0.01)。与空白对照组相比,复方透骨草酊组及米诺地尔酊组图像和凋亡指数均无明显差异(P> 0.05)。4组在血清雌二醇水平上无统计学差异(P> 0.05)。结论 复方透骨草酊能够促进毛发生长,其作用机制可能与缩短毛发生长休止期、延长生长期时间,降低血清中睾酮、双氢睾酮水平,抑制毛囊细胞凋亡相关。

Objective To investigate the effect of compound Tuogucao tincture on sex hormone levels,hair growth and hair follicle cell apoptosis in C57BL/6 alopecia model mice induced by testosterone.Methods 40 mice were randomly divided into four groups:blank control group,model group,minoxidil group and Tougucao tincture group.The mice were given external treatment in the hairless area and killed after collecting blood by eyeball enucleation.Blood samples were used to detected the levels of serum testosterone,estradiol and dihydrotestosterone.The apoptosis level of hair follicle cell was detected by TUNEL red fluorescence assay.Results Compared with the model group,there were significant statistical differences in hair growth rate,serum testosterone and dihydrotestosterone levels,and apoptosis index in the Tougucao tincture group(P<0.01).Compared with the blank control group,there was no significant difference in the image and the apoptotic index of the Tougucao tincture group and the minoxidil group(P>0.05).There was no statistical difference in the serum estradiol levels among the four groups(P>0.05).Conclusion The compound Tougucao tincture can promote hair growth,and the mechanism of action may be related to shortening the resting period of hair growth,lengthening the growth period,reducing the level of serum testosterone and dihydrotestosterone,and inhibiting the apoptosis of hair follicle cells.
