摘要
本文建立了绿豆糕中脱氢乙酸的高效液相色谱法快速测定方法,并采用测定绿豆糕样品加标回收率的方法对检测方法进行验证;同时探讨了流动相乙酸铵的pH值变化对脱氢乙酸峰形的影响。结果显示脱氢乙酸浓度与响应值之间的线性关系良好(R^(2)=0.9999),样品的加标回收率为95%~100%,相对标准偏差为1.2%~1.7%;在流动相甲醇∶乙酸铵=7∶93的条件下,乙酸铵pH值调节至8.5时,脱氢乙酸的出峰效果好,峰形尖锐且对称。该方法准确度高,且具有很好的重现性,适用范围广,简单易操作,实用性强。
In this paper,a rapid determination method of dehydroacetic acid in mung bean cake was established by high performance liquid chromatography,and the method of measuring the recovery rate of mung bean cake sample was used to verify the detection method.At the same time,the effect of pH change of mobile phase ammonium acetate on the peak shape of dehydroacetic acid was discussed.The results showed that the linear relationship between the concentration of dehydroacetic acid and the response value was good(R^(2)=0.9999),the recovery rates of the samples were 95%~100%,and the relative standard deviations were 1.2%~1.7%;under the condition of mobile phase methanol∶ammonium acetate=7∶93,when the pH value of ammonium acetate is adjusted to 8.5,the peak effect of dehydroacetic acid is good,and the peak shape is sharp and symmetrical.The method has high accuracy,good reproducibility,wide application range,simple operation and strong practicability.
