微生物群基因编辑技术演进分析

摘要 目的:描述规律成簇的间隔短回文重复(Clustered Regularly Interspaced Short Palindromic Repeats,CRISPR)领域技术演化,揭示微生物群基因编辑技术的演进路线,对其技术发展进行深层分析,以期为相关领域的研究人员提供决策参考。方法:计算机检索PubMed和Embase数据库截至2021年5月30日收录的相关文献,筛选微生物群基因编辑技术相关研究。结果:初步检索902篇文献,最终44篇符合纳入标准;研究显示CRISPR技术演进方向主要包括特异性、适应性和多样性,其中60.81%涉及多样性研究、28.38%涉及适应性研究、10.81%涉及特异性研究;围绕基因编辑技术整体研究重点不一,以辅助系统的多样性技术为主。结论:下一代测序技术及人工智能、大数据的发展助力微生物群基因编辑技术研究趋向多元化与细致化,并为其提供了新方向。 Objective: To study and describe the technological evolution in Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR) field, reveal the evolution route of microbiota gene editing technology, and make an indepth analysis of its technological development, so as to provide reference for researchers in related fields. Methods:The relevant literatures collected in PubMed and Embase databases before May 30, 2021 were searched by computer to screen the related research of microbiota gene editing technology. Results: 902 literatures were initially searched,and 44 of them met the inclusion criteria. The research showed that the evolution direction of CRISPR technology mainly included specificity, adaptability and diversity, of which 60.81% involved diversity research, 28.38% involved adaptability research and 10.81% involved specificity research;The development of auxiliary tools is mainly based on diversity, and the overall research focus is different around gene editing technology, mainly based on the diversity technology of auxiliary system. Conclusion: The development of next-generation sequencing technology, artificial intelligence and big data contributes to the diversification and refinement of microbiota gene editing technology, and provides a new trend for it.
