摘要 目的:探讨研制的胱抑素C试剂在Hitachi 7180和Beckman Coulter AU680全自动生化分析仪上的钩状效应差异及其解决方法。方法:以免疫比浊法测定胱抑素C为研究对象,通过配制一系列不同浓度梯度的胱抑素C抗原溶液,在两种生化分析仪上进行检测,对比分析其钩状效应差异,并研究通过调整抗体用量解决在两种分析仪上的钩状效应差异。结果:当抗体用量为100 mg/L时,在Hitachi 7180全自动生化分析仪上检测出现钩状效应的抗原浓度为14 mg/L左右,在Beckman Coulter AU680全自动生化分析仪上检测出现钩状效应的抗原浓度为12 mg/L左右,两者的差异最小,对临床检测的也影响最小。结论:所研制的胱抑素C试剂应充分考虑在不同仪器上检测的钩状效应差异,以此指导临床检测过程中对抗原超限标本进行稀释处理,从而避免在检测过程中由于检测仪器的不同而造成超高值标本检测结果的不准确。 Objective: To investigate the difference of hook effect of cystatin C reagent on Hitachi 7180 automated chemistry analyzer and Beckman Coulter AU680 automated chemistry analyzer and the solution. Methods: The immunoturbidimetry assay for the determination of cystatin C was taken as the research object, a series of concentration gradient cystatin C antigen solutions were prepared and tested to compare the difference of hook effect on different automated chemistry analyzers, and whether adjusting the concentration of cystatin C antibody could solve the hook effect was also researched. Results: When the concentration of antibody is 100 mg/L, the hook effect occurred at the approximate antigen concentration of 14 mg/L on Hitachi 7180 automated chemistry analyzer and 12 mg/L on Beckman Coulter AU680 automated chemistry analyzer. The difference between them is the smallest and had the smallest impact on clinical detection. Conclusion: The research of cystatin C reagent should fully consider the difference of hook effect on different automated chemistry analyzer, so as to guide the dilution of high-concentration antigen samples in the process of clinical detection, so as to avoid the inaccuracy of detection results of ultra-high-concentration antigen samples due to different detection instruments.
