摘要
本文建立了高效液相色谱法测定婴幼儿配方奶粉中乳铁蛋白的方法。样品用磷酸盐缓冲液提取,经肝素亲和柱净化后,C_(4)反相色谱柱分离,DAD检测器于280 nm处检测,用外标法定量。结果表明,乳铁蛋白在浓度10~200 mg/L范围内曲线的线性关系良好,方法定量限为15 mg/100 g,检出限为5 mg/100 g,3个添加水平(150 mg/kg、300 mg/kg、1000 mg/kg)的平均回收率为81.0%~92.0%,相对标准偏差在1.65%~3.92%之间。该方法操作方便、简单、重现性好,适用于婴幼儿配方奶粉中乳铁蛋白的测定。
The High Performance Liquid Chromatography method(HPLC)was established for determining lactoferrin in infant formula.The sample was extracted by phosphate buffer,purified by heparin affinity column,separated by C_(4) reversed-phase column,and detected by DAD detector at UV 280 nm.The results showed that lactoferrin had a good linear coefficient in the concentration range of 10-200 mg/L.The limit of quantitation was 15 mg/100 g,and the limit of detection was 5 mg/100 g.The average recoveries of 3 spiked level(150 mg/kg,300 mg/kg,1000 mg/kg)were between 81.0%-92.0%and relative standard deviations were between 1.65%-3.92%.This method is convenient,accurate and reproducible,thus suitable for determination of lactoferrin in infant formula.
