甘薯近缘野生资源的染色体倍性研究

摘要 以甘薯近缘野生种Ipomoea trifida根尖为材料,分析0.1%秋水仙素、对二氯苯饱和溶液、2 mmol/L的8-羟基喹啉三种试剂不同处理时间对细胞有丝分裂指数、制片效果的影响。结果表明,对二氯苯饱和溶液对甘薯野生种I.trifida根尖进行3 h处理,细胞有丝分裂指数最高,染色体制片效果较好,为最佳预处理方法。基于上述体系鉴定了本试验中的甘薯野生种I.trifida为二倍体(2 n=2 x=30),秋水仙素加倍处理后的I.trifid是四倍体(2 n=4 x=60),徐薯18是六倍体(2 n=6 x=90)。 Using the root tips of the closely related wild species Ipomoea trifida as materials.The three reagents including 0.1%colchicine,p-dichlorobenzene saturated solution and 2 mmol/L 8-hydroxyquinoline were used to analyze the effects on slicing of root tip in different treatment time,the root tips of Ipomoea trifida from a wild relative species of sweet potato.The highest mitotic index and the optimal slide specimen were obtained when root tips were treated with p-dichlorobenzene saturated solution for 3 hours.Based on the above system,the wild species of sweet potato I.trifida was idendified as diploid,I.trifida identified as tetraploid after colchicine doubling,Xinshu 18 identified as hexaploid.
