圆锥绣球“粉色精灵”组培快繁技术研究

Study on Tissue Culture and Rapid Propagation ofHydrangea paniculata ‘Pink Winky’

以“粉色精灵”幼嫩的茎段为外植体,探索圆锥绣球的组培快繁技术。结果表明,将外植体在洗衣液中浸泡15 min,流水下冲洗1.5~2 h,在2 g/L多菌灵和0.6 g/L链霉素的混合液中浸泡8 min,再在超净工作台内用75%酒精浸泡40 s,0.1%HgCl_(2)浸泡5 min,灭菌效果最好,成活率达95%。启动培养配方为:MS+6-BA 0.5 mg/L+NAA 0.15 mg/L,萌芽率达100%。适宜的增殖培养配方为:MS+6-BA(1.0~1.5)mg/L+NAA(0.1~0.15)mg/L,增殖系数最高可达7.0。最适生根培养基为1/2 MS+6-BA 0.05 mg/L+IBA 0.1 mg/L+NAA 0.1 mg/L,生根率达100%,平均根数21条,平均根长3.12 cm。适宜的移栽基质为V_(腐殖土)∶V_(珍珠岩)=1∶1的混合基质,移栽成活率最高为87%。

In this study,the young stem segment of Hydrangea paniculata‘Pink Winky’was used as explants,The tissue culture and rapid propagation techniques were tested.The results showed that the explants were washed under running water for 1.5-2 h,soaked in washing liquid for 15 min,soaked in 2 g/L Carbendazim and 0.6 g/L streptomycin mixture for 8 min,and then the stem segments were disinfected in ultra clean worktable,soaked in 75%alcohol for 40 s,soaked in 0.1%mercuric chloride for 5 min,and the bactericidal effect was the best,the survival rate reaching 95%.The starter medium formula was MS+6-BA 0.5 mg/L+NAA 0.15 mg/L,the germination rate reached 100%.The optimal formula for proliferation culture was MS+6-BA(1.0-1.5)mg/L+NAA(0.1-0.15)mg/L,the proliferation coefficient was up to 7.0.The optimum rooting medium was 1/2 MS+6-BA 0.05 mg/L+IBA 0.1 mg/L,under which conditions the rooting rate was 100%,and the average root number was 21,and the average root length was 3.12 cm.The suitable transplanting medium was the mixture of humus and perlite with the volume ratio of 1∶1,and the highest transplanting survival rate was 87%.
