淡竹叶标准汤剂UPLC特征图谱研究

Study on UPLC Characteristic Spectrum of Standard Decoction of Lophatherum Gracile Brongn

目的:建立淡竹叶药材与标准汤剂超高效液相色谱(UPLC)特征图谱,对其特征图谱的相关性进行分析评价。方法:参照标准汤剂制备方法,制备17批淡竹叶标准汤剂,采用Waters CORTECS T3色谱柱,以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱,流速为0.25mL/min,柱温为40℃,检测波长为350nm,采用中药色谱指纹图谱相似度评价系统软件进行相似度分析。结果:17批淡竹叶药材、标准汤剂共标定出共有特征峰4个,其中峰2为异荭草苷,峰3为日当药黄素,分离度较好,药材和标准汤剂与各自参照图谱呈良好的相关性。结论:建立了淡竹叶药材、标准汤剂的UPLC特征图谱,特征图谱相似度较高,可以全面反映淡竹叶药材、标准汤剂的整体信息,通过特征图谱共有峰,可辨别混伪品,从源头监控淡竹叶的质量,为淡竹叶中药配方颗粒的全过程控制提供了参考依据。

Objective:To establish the UPLC characteristic spectra of raw herb and standard decoction of Lophatherum gracile Brongn.and to analyze the correlation of their characteristic spectra.Methods:17 batches of Lophatherum gracileBrongn.standard decoctions were prepared following the preparation conditions of the standard decoction.UPLC system was performed with the Waters CORTECS T3 column(2.1mm×100mm,1.6μm),gradient mobile phase of acetonitrile-0.1% phosphoric acid solution,flow rate of 0.25mL/min and column temperature of 40℃,detective wavelength of 350 nm.The chromatographic characteristic spectrum were analyzed with the similarity evaluation system for chromatographic characteristic spectrum of Traditional Chinese medicine.Results:There were 4 common characteristic peaks in all characteristic spectrum of 17 batches of raw herbs and standard decoctions of Lophatherum gracile Brongn.,showed good correlation and separate well on the whole.Among them,peak 2 was isoorientin,and peak 3 was flavin.Conclusion:To establish the UPLC characteristic spectra of raw herb and standard decoction of Lophatherum gracile Brongn,which fully reflected the overall situation of raw herb and standard decoction of Lophatherum gracile Brongn.There was a high similarity and the common peaks of the characteristic spectra could be used for the identification of fake.The quality of Lophatherum gracile Brongn.provides a method for the whole process control of Lophatherum gracile Brongn.Chinese medicine formula granules.
