摘要
目的:运用网络药理学对金铃子散治疗乳腺癌(BC)的机理进行数据挖掘和分析。方法:通过TCMSP数据库选出金铃子散活性成分的对应靶点,与Gene Cards网站选取的乳腺癌靶点取交集后获得潜在靶标基因。应用Cytoscape软件构建金铃子散-活性成分-靶点-乳腺癌可视化网络图;应用STRING在线软件展示蛋白-蛋白间的相互作用关系,并应用Cyto NCA插件进行度中心性分析得到核心靶点。使用DAVID在线工具和R计算机语言进行GO和KEGG注释。结果:金铃子散治疗乳腺癌的共同靶标蛋白有107个,其中核心靶标蛋白包括蛋白激酶B(AKT1)、雌激素受体(ESR1)、血管内皮生长因子A(VEGFA)等。GO分析得到BP生物过程,MF分子功能和CC细胞功能。KEGG通路分析显示40个条目,涉及与肿瘤疾病类相关通路、炎症有关通路、感染相关疾病类通路、信号转导通路等。结论:金铃子散通过核心靶点参与了一系列通路,通过发挥分子功能及多种生物过程防治乳腺癌,其中PI3K/AK信号通路很可能是其中的关键通路。
Objective:To explore he mechanism of Jinlingzi powder for breast cancer by usingnetwork pharmacology for data mining.Methods:The active components and corresponding targets of Jinlingzi powder retrieved from the TCMSP database,combining with breast cancer genes obtained from GeneCards so that overlapping potential targets were kept.Using the Cytoscape Software to construct the visualization diagram of Jinlingzi Powder-active ingredient-targetbreast cancer.The STRING online software was used to show the protein-protein interaction,and CytoNCA plug-in was used for degree centrality analysis to obtain the core targets.GO and KEGG annotations,are performed using the DAVID online tool and the R computer language.Results:There are 107common target proteins of Jinlingzi powder for the treatment of breast cancer,among which the core targets include protein kinase B(AKT1),estrogen receptor(ESR1),and vascular endothelial growth factor A(VEGFA),etc.The biological process of BP,molecular function of MF and cell function of CC were obtained by GO analysis.KEGG pathway analysis showed 40entries,involving pathways related to tumour diseases,inflammation-related pathways,infection-related diseases,signal transduction pathways,etc.Conclusion:Through the core target,Jinlingzi powder participates in a series of pathways to exert molecular functions and a variety of biological processes to prevent and treat the breast cancer,among which the PI3K/AKT signalling pathway is likely to be the critical pathway.
