冠心病患者血清miR-144和FOXF1表达及与冠状动脉病变严重程度的关系 被引量:2

Correlation between the expressions of serum miR-144,FOXF1 and the severity of coronary artery lesion in patients with coronary heart disease

摘要 目的探讨冠心病患者血清miR-144和叉头盒蛋白F1(FOXF1)表达与冠状动脉病变严重程度的关系。方法选择2018年1月至2020年3月收治的108例冠心病患者(冠心病组)和100例年龄、性别相匹配的体检志愿者(对照组)。采用RT-PCR法检测2组受试者miR-144和FOXF1表达,冠心病组行冠状动脉造影术评估冠脉病变支数、冠脉狭窄程度和冠脉病变程度。比较不同冠脉病变患者血清miR-144和FOXF1表达差异,Pearson或Spearman秩相关分析miR-144和FOXF1相关性以及miR-144、FOXF1和冠脉病变支数、冠脉狭窄率、SYNTAX积分的相关性。受试者工作特征曲线(ROC)分析miR-144、FOXF1诊断重度病变的价值。结果冠心病组血清miR-144表达高于对照组(P<0.05),FOXF1表达低于对照组(P<0.05)。血清miR-144表达随着冠脉病变支数增加,冠脉病变程度加重而增高(P<0.05),FOXF1则降低(P<0.05),冠脉狭窄Ⅲ度~Ⅳ度组血清miR-144表达高于Ⅰ度~Ⅱ度组(P<0.05),FOXF1表达低于Ⅰ度~Ⅱ度组(P<0.05)。miR-144表达与冠脉病变支数、冠脉狭窄率、SYNTAX积分呈正相关(r/rs值分别为0.745、0.812、0.842,P<0.05),FOXF1表达与冠脉病变支数、冠脉狭窄率、SYNTAX积分呈负相关(r/rs值分别为-0.739、-0.806、-0.859,P<0.05),miR-144表达与FOXF1表达呈负相关(r=-0.927,P<0.05)。联合miR-144、FOXF1诊断重度病变的曲线下面积为0.932,高于单独miR-144、FOXF1(Z=2.412、2.569,P<0.05)。结论冠心病患者血清miR-144表达上调,FOXF1表达下调,miR-144、FOXF1均与冠脉病变程度有关,miR-144可能通过负性调控FOXF1表达参与冠脉病变过程。检测miR-144、FOXF1有助于鉴别冠脉病变程度。 Objective To investigate the correlation between the expressions of serum miR-144,FOXF1 and the severity of coronary artery lesion in patients with coronary heart disease(CHD).Methods A total of 108 patients with CHD(CHD group)and 100 volunteers with the same age and gender as those in CHD group who underwent physical examination(control group)from January 2018 to March 2020 were enrolled in the study.The expression levels of miR-144 and FOXF1 were detected by RT-PCR,and the number of coronary artery lesions,the degree of coronary artery stenosis and the degree of coronary artery lesions were assessed by coronary angiography in the CHD group.And the expression levels of miR-144 and FOXF1 were observed in patients with different coronary artery diseases.Moreover the correlation between miR-144 and FOXF1,as well as the correlation between miR-144,FOXF1 and the number of coronary lesions,coronary stenosis rate and SYNTAX score were analyzed by Pearson or Spearman rank correlation.Results The expression levels of serum miR-144 in CHD group were significantly higher than those in control group(P<0.05),however,the expression levels of FOXF1 significantly lower than those in control group(P<0.05).The expression levels of serum miR-144 were increased with the increase of the number of coronary artery lesions and the aggravation of coronary artery lesions(P<0.05),while FOXF1 levels were significantly decreased(P<0.05).The expression levels of miR-144 inⅢ~Ⅳgrade group were significantly higher than those inⅠ~Ⅱgrade group(P<0.05),but,the expression levels of FOXF1 were significantly lower than those inⅠ~Ⅱgrade group(P<0.05).The expression levels of miR-144 were positively correlated with the number of coronary lesions,the rate of coronary stenosis,and SYNTAX score(P<0.05),however,the expression levels of FOXF1 were negatively correlated with these parameters(P<0.05),and the expression of miR-144 was negatively correlated with that of FOXF1(P<0.05).The area under the curve of combined miR-144 and FOXF1 in diagnosis of severe lesions was significantly higher than that by simple index detection(P<0.05).Conclusion The expression levels of serum miR-144 are up-regulated and the expression levels of FOXF1 are down-regulated in patients with CHD.Both miR-144 and FOXF1 are related to the degree of coronary artery disease,and miR-144 may be involved in the process of CHD by negatively regulating the expression of FOXF1,and to detect the expression of miR-144 and FOXF1 in helpful to identify the degree of coronary artery lesions.
