IFIT5在子宫内膜癌组织中的表达及其对内膜癌HEC-1-A细胞生物学行为的影响

摘要 目的探究干扰素诱导蛋白(interferon-induced protein with tetratricopeptide repeats 5,IFIT5)在子宫内膜癌组织中的表达及其对子宫内膜腺癌细胞HEC-1-A生物学行为的影响。方法采用免疫组织化学染色法检测IFIT5在30例正常内膜组织及60例子宫内膜癌组织中的表达,并分析其过表达与子宫内膜癌临床病理特征的相关性;采用脂质体转染法将订制的特异siRNA转染至内膜癌细胞内,靶向抑制IFIT5的表达,并通过CCK-8法、Transwell小室、划痕实验检测干扰前后HEC-1-A细胞增殖、侵袭、迁移能力变化。结果与正常子宫内膜组织比较,IFIT5在子宫内膜癌组织中表达较高(P<0.05),且IFIT5的高表达与子宫内膜癌的临床分期、肌层浸润深度、淋巴结转移情况等相关(P<0.05),但与年龄无关(P>0.05);siRNA抑制IFIT5表达后,CCK-8、Transwell小室及划痕实验显示,抑制组细胞的增殖、侵袭及迁移能力均较阴性对照组显著降低(P<0.05)。结论IFIT5在子宫内膜癌组织中呈现高表达,其高表达与子宫内膜癌的发生、发展相关;抑制IFIT5的表达可抑制子宫内膜癌细胞的增殖、迁移和侵袭。 Objective To explore the expression and clinical significance of interferon-induced protein with tetratricopeptide repeats 5(IFIT5)in endometrial carcinoma and its effect on the biological behavior of HEC-1-A endometrial adenocarcinoma cells.Methods Immunohistochemical staining was used to detect the expression of IFIT5 in 30 cases of normal endometrial tissues and 60 cases of endometrial carcinoma tissues,and the correlation between its overexpression and the clinicopathological features of endometrial carcinoma was analyzed.Liposome transfection method was used to transfect specific siRNA into endometrial cancer cells,and the expression of IFIT5 was targeted to inhibit.The changes in the proliferation,invasion and migration of HEC-1-A cells before and after interference were detected by CCK-8 method,Transwell chamber and scratch test.Results Compared with normal endometrial tissues,the expression of IFIT5 was higher in endometrial carcinoma tissues(P<0.05).The high expression of IFIT5 was correlated with the clinical stage of endometrial carcinoma,the depth of muscular invasion and lymph node metastasis(P<0.05),but not with age(P>0.05).After siRNA inhibiting the expression of IFIT5,CCK-8,Transwell and scratch test showed that the proliferation,invasion and migration of cells in the inhibition group were significantly lower than those in the negative control group(P<0.05).Conclusion IFIT5 is highly expressed in endometrial cancer tissues,and its high expression is related to the occurrence and development of endometrial cancer.Inhibition of IFIT5 expression can inhibit the proliferation,migration and invasion of endometrial cancer cells.
