摘要
目的:建立猫头刺HPLC指纹图谱及3个活性成分的含量测定方法。方法:采用HPLC法,Eclipse Plus C_(18)色谱柱(250 mm×4.6 mm,5.0μm);以乙腈(A)-0.1%乙酸水溶液(B)梯度洗脱;流速为1.0 mL·min^(-1);柱温30℃;检测波长330 nm。结果:建立了猫头刺HPLC指纹图谱,共标定27个共有峰,指认了其中6个共有峰;同时测定17批次药材中二氢山柰酚、圣草酚和木犀草素的含量分别为0.0296~2.7127 mg·g^(-1),0.0566~0.5287 mg·g^(-1),0.0213~0.8321 mg·g^(-1)。结论:建立的指纹图谱结合3种成分的含量测定方法稳定可靠,可为猫头刺药材质量评价提供参考。
OBJECTIVE To establish the HPLC fingerprint of Oxytropis aciphylla and content determination method for three active components.METHODS The HPLC method was performed at 30℃on an Eclipse Plus C_(18) column(250 mm×4.6 mm,5.0μm),eluted with a gradient program by using acetonitrile(A)-0.1%acetic acid aqueous solution(B)as the mobile phase.The flow rate was 1.0 mL·min^(-1),and the detection wavelength was 330 nm.RESULTS The HPLC fingerprint of Oxytropis aciphylla was established.A total of 27 common peaks were calibrated and six common peaks were identified.At the same time,the contents of dihydrokaempferol,eriodictyol and luteolin in 17 batches of samples were determined,which were 0.0296-2.7127 mg·g^(-1),0.0566-0.5287 mg·g^(-1) and 0.0213-0.8321 mg·g^(-1),respectively.CONCLUSION The established fingerprint combined with the content determination method for the three components is stable and reliable,which provides reference for the quality evaluation of Oxytropis aciphylla.
