基于牡荆素的绿豆大曲质量控制方法研究

Research on quality control method of Lvdou Daqu based on Vitexin

目的:选择绿豆中主要的黄酮类活性成分牡荆素为养生酒绿豆大曲的质量控制指标,制订质量控制标准。方法:牡荆素的分析采用Infinity Lab Poroshell 120 EC-C18(4.6 mm×100 mm,2.7 Micron)色谱柱;流动相为乙腈-超纯水,梯度洗脱;体积流量0.3 mL·min^(-1),柱温40℃。建立HPLC-UVD检测绿豆大曲中功效指标成分方法,测定两类绿豆大曲样品中功效指标成分含量,根据定量结果建立质控标准。结果:两类绿豆大曲中牡荆素含量有显著差异,分开制订质量控制标准,A类为0.03mg·mL^(-1),B类为0.05mg·mL^(-1)。结论:保健酒中来源于功效添加原料的主要功效成分可作为质控指标,为对产品功效添加的一致性提供定性定量控制方法。

Objective:The main flavonoid active ingredient vitexin in Lvdou was selected as the quality control index of health wine Lvdou Daqu,and the quality control standard was formulated.Method:Vitexin was analyzed using InfinityLab Poroshell 120 EC-C18(4.6 mm×100 mm,2.7 Micron)chromatographic column.Mobile phase was acetonitrile-ultrapure water in gradient elution with volume flow rate of 0.3 mL·min^(-1)at 40℃.An HPLC-UVD method was established for detecting the effcacy index components in Lvdou Daqu,and detecting the contents of the efficacy index components in the two types of Lvdou Daqu samples.The quality control standard was established based on the quantitative results.Result:Vitexin,the main flavonoid from Lvdou Daqu,was selected as the quality control index.The content of vitexin in the two types of Lvdou Daqu was significantly different.The quality control standards were formulated separately.Class A was 0.03 mg·mL^(-1),and class Bwas 0.05 mg·mL^(-1).Conclusion:The main functional ingredients in health wines derived from functional additives can be used as quality control indicators to provide a qualitative and quantitative control method for the consistency of product functional additions.
