魔芋醇提取物急性毒性试验及初步药效作用研究

目的:考察魔芋醇提取物的急性毒性和药理作用研究。方法:采用单次灌胃给药考察魔芋醇提取物的急性毒性,采用小鼠肠推进实验考察魔芋醇提取物的促消化和润肠通便作用;采用S180和H22小鼠抑瘤实验考察魔芋醇提取物的抗肿瘤活性。结果:急性毒性试验,魔芋醇提物24 h内小鼠灌胃给药LD_(50)为19.18 g·kg^(-1)(相当于生药165.34 g·kg^(-1));小鼠肠推进实验,与空白对照组比较,魔芋醇提取物低剂量组和高剂量组小鼠小肠的墨汁推进百分率均显著升高(均P<0.01),抗肿瘤作用研究,与空白对照组比较,魔芋醇提取物低剂量组和高剂量组可以显著降低小鼠瘤重(P<0.05或P<0.01);魔芋醇提取物高剂量组对H22移植瘤小鼠的胸腺指数和脾脏指数均显著增加(P<0.05或P<0.01)。结论:魔芋醇提取物具有显著的促消化,润肠通便的功效;可以抑制S180和H22肿瘤细胞的生长,具有显著的抗肿瘤活性;且具有一定的毒性。

Objective:To investigate the acute toxicity and the pharmacological effects of Moyu alcohol extract.Method:A single intragastric administration was used to investigate the acute toxicity.The mouse intestinal propulsion experiment was used to investigate the digestive and intestinal laxative effects,and the S180 and H22 mouse tumor suppression experiments were used to investigate the anti-tumor activity.Result:In the acute toxicity test,the LD_(50) of Moyu alcohol extract administered intragastrically to mice within 24h was 19.18 g·kg^(-1)(equivalent to 165.34 g·kg^(-1)of crude drugs).In the mouse intestinal propulsion test,compared with the control group,the low-dose and high-dose Moyu alcohol extract groups significantly increased the bowel advancement distance(both P<0.01).In the anti-tumor effect study,compared with the control group,the low-dose and high-dose Moyu alcohol extract groups significantly reduced mouse tumor weight(P<0.05 or P<0.01).The high-dose Moyu alcohol extract group significantly increased the thymus index and spleen index of H22 transplanted mice(P<0.05 or P<0.01).Conclusion:Moyu alcohol extract has significant digestion-promoting and laxative effects.It can inhibit the growth of S180 and H22 tumor cells and has significant anti-tumor activity,but it also has some toxicity.
