人外周血单核细胞抵抗鼻病毒感染的机制研究 被引量:1

Gene changes and mechanism of rhinovirus infection peripheral blood mononuclear cells

摘要 目的分析鼻病毒在感染人外周血单核细胞后发生的基因表达变化,并分析人外周血单核细胞应对鼻病毒感染的过程及机制,探索鼻病毒感染人外周血单核细胞后启动的免疫反应。方法从GEO数据库通过限定词搜索与鼻病毒及人外周血单核细胞相关的数据集,限定词为:rhinovirus;mononuclear cells;series;homo sapiens;expression profiling by array。经过筛选找到数据集GSE53543,下载GSE53543数据集的文件资料,分析感染与未感染鼻病毒的人外周血单核细胞中的基因表达差异,进行GO/KEGG聚类分析、免疫检测点的相关性分析及铁死亡相关基因的分析。结果按照假定的条件,共发现有281个基因存在表达差异,结果包括217个上调的基因,64个基因表达下调;GO富集分析后富集数最多的是与感染相关及抵御感染有关的途径;KEGG的通路富集中,以参与NOD样受体信号的通路最多;免疫相关性分析共找到4个有差异的免疫基因;铁死亡相关分析找到了14个与铁死亡相关的基因。结论鼻病毒感染后会导致人外周血单核细胞的基因发生显著变化,通过GO/KEGG富集分析及免疫检测点、铁死亡的分析,预测了鼻病毒感染人体后的发病机制以及外周血单核细胞在鼻病毒感染后启动了免疫反应。 Objective To analyze the gene expression changes of human peripheral blood mononuclear cells after rhinovirus infection,and to analyze the mechanism of human peripheral blood mononuclear cells in response to rhinovirus infection,to explore the immune response of human peripheral blood mononuclear cells to rhinovirus infection.Methods Search from the GEO to find the profile data,use criteria:rhinovirus;mononuclear cells;series;homo sapiens;expression profiling by array.Then find the series of GSE53543.GEO2 R was used to study the differential expression of Genes.Perform GO/KEGG cluster analysis,immunoassay and iron death analysis.Results Totally 281 DEGS were found,including 217 high(logFC positive)expressions and 64 low(logFC negative)expressions.GO analysis showed that the most enrichment was related to infection and anti-infection pathway.KEGG analysis showed that the most enrichment was in NOD receptor signaling pathway.The immune correlation analysis found four different immune genes.Iron death correlation analysis found 14 iron death related genes.Conclusion The genes of human peripheral blood mononuclear cell were significant changed after rhinovirus infection.The mechanism of rhinovirus infection was predicted by functional analysis and immunoassay and iron death.Predicte the pathogenesis of rhinovirus infection,and rhinovirus initiate the immune response of peripheral blood mononuclear cells.
