黄连生物碱改善心力衰竭的网络药理学机制研究 被引量:2

Study on the Mechanism of Coptis Alkaloid in the Treatment of Heart Failure Based on Network Pharmacology

摘要 目的:应用网络药理学探讨黄连生物碱治疗心力衰竭的机制。方法:对黄连生物碱进行中药系统药理学数据库与分析平台(traditional Chinese medicine database and analysis platform,TCMSP)检索,通过通用蛋白质资源平台(the universal protein resource,UniProt)数据库筛选靶点;通过治疗靶点数据库(therapeutic target database,TTD)、人类基因数据库(the human gene database,GENECARDS)数据库检索心衰靶点,通过venny交集,获取重叠靶点应用String数据库、Cytoscape 3.7.1绘制蛋白-蛋白作用网络及分析参数,通过Metascape数据库进行富集分析。结果:富集分析KEGG通路,筛选出5条直接与心力衰竭相关的信号通路,主要为神经活性配体-受体通路、钙信号通路、cGMP-PKG信号通路、HIF-1信号通路、cAMP信号通路。结论:黄连生物碱治疗心力衰竭的作用机制主要与神经体液、离子信号通路等相关。 Objective:To discuss the mechanism of coptis alkaloid in the treatment of heart failure by using network pharmacology.Methods:Coptis alkaloid was searched from TCMSP,and the targets were screened via UniProt;the targets of heart failure were retrieved from TTD and GENECARDS,the overlapping targets were obtained by venny intersection,protein protein interaction network was drawn and the parameters were analyzed by applying String database,Cytoscape 3.7.1,enrichment analysis was performed by using Metascape database.Results:Through enrichment analysis KEGG pathways,there are five signaling pathway related to heart failure directly,including neuroactive ligand receptor interaction,calcium ion channels,cGMP-PKG signaling pathway,HIF-1 signaling pathway and cAMP signaling pathway.Conclusion:The mechanism of coptis alkaloid in the treatment of heart failure is primarily related to neurohumoral and ionic signaling pathways.
