Expression of SMARCA4 in hepatocellular carcinoma and its clinical significance

目的研究SMARCA4在肝癌组织中的表达水平,并分析其临床意义。方法选取于广西医科大学附属肿瘤医院肝胆外科2018年1~12月进行手术并经病理证实为原发性肝细胞型肝癌的40例患者为研究对象,采用实时荧光定量PCR、Western blot法检测患者肝癌组织及癌旁组织中SMARCA4的表达水平;采用t检验分析SMARCA4蛋白与肝癌患者临床病理特征的关系。结果肝癌组织中SMARCA4 mRNA的表达水平为6.36±0.57,明显高于癌旁组织的1.31±0.31,差异有统计学意义(P<0.05);肝癌组织中SMARCA4蛋白的表达水平为0.96±0.33,明显高于癌旁组织的0.45±0.14,差异有统计学意义(P<0.05);女性肝癌患者SMARCA4的表达水平明显高于男性,甲胎蛋白(AFP)大于2000 ng/mL的肝癌患者SMARCA4的表达水平明显高于AFP小于2000 ng/mL的肝癌患者,差异均有统计学意义(P<0.05)。结论SMARCA4在肝癌组织中高表达,且与患者性别和AFP水平可能相关,具有一定的临床参考价值。

Objective To study the expression level of SMARCA4 in hepatocellular carcinoma and to analyze its clinical significance.Methods Forty patients with primary hepatocellular carcinoma confirmed by pathology were selected from Department of Hepatobiliary Surgery,Cancer Hospital Affiliated to Guangxi Medical University from January 2018 to December 2018.Real-time fluorescence quantitative PCR and Western blot were used to detect the expression level of SMARCA4 in hepatocellular carcinoma and adjacent tissues,and t-test was used to analyze the relationship between SMARCA4 protein and clinicopathological features of patients with hepatocellular carcinoma.Results The expression level of SMARCA4 mRNA in HCC tissues was 6.36±0.57,significantly higher than 1.31±0.31 in adjacent tissues(P<0.05),and the expression level of SMARCA4 protein in HCC tissues was 0.96±0.33,significantly higher than 0.45±0.14 in adjacent tissues(P<0.05).The expression level of SMARCA4 in female patients with liver cancer was significantly higher than that in males(P<0.05).The expression level of SMARCA4 in patients with AFP>2000 ng/mL was significantly higher than that in patients with AFP<2000 ng/mL.Conclusion SMARCA4 is highly expressed in HCC tissues,and may be related to the sex of patients and the level of AFP,which has a certain clinical reference value.
