摘要
目的优化超声法提取玛咖多糖的工艺,并初步研究玛咖多糖对大鼠小肠上皮细胞(IEC-6)的增殖及其迁移的作用。方法以水为溶剂,以玛咖多糖得率作为响应指标,以液料比(体积∶质量)、超声功率、超声时间以及提取温度为主要因素,采用单因素实验结合响应面分析实验,优化工艺。分别采用MTT法、细胞划痕法,初步研究玛咖多糖对IEC-6细胞增殖以及迁移的作用。结果当提取时间83min,提取功率177W、液料比40ml/g、提取温度63℃时,多糖得率最高,平均得率是(75.20±1.21)mg/g。玛咖多糖浓度在0.1~10mg/L范围内,能够促进IEC-6细胞增殖,在10mg/L浓度时玛咖多糖促进IEC-6细胞迁移。结论通过响应面优化的超声法提取的玛咖多糖,在一定浓度条件下可以促进IEC-6细胞的增殖以及迁移。
Objective To optimize the ultrasonic extraction process for extracting Maca polysaccharide,and to study the effect of Maca polysaccharide on the proliferation and migration of small intestinal epithelial cells(IEC-6)in rats.Methods With water as the solvent,Maca polysaccharide yield as the response index,and liquid-solid ratio(volume:mass),ultrasonic power,ultrasonic time and extraction temperature as the main factors,a single factor experiment combined with response surface analysis experiment was performed to optimize the process.MTT assay and cell scratch assay were adopted to observe the effect of Maca polysaccharide on the proliferation and migration of IEC-6cell respectively.Results When the extraction duration was83min,the extraction power was 177W;the liquid-solid ratio 40ml/g.When the extraction temperature was63℃,the polysaccharide yield was the highest,with an average yield of(75.20±1.21)mg/g.At the concentration of 0.1~10mg/L,Maca polysaccharide could promote the proliferation of IEC-6cells.At the concentration of 10mg/L,Maca polysaccharide could promote the migration of IEC-6cells.Conclusion Maca polysaccharide extracted by response surface optimized ultrasonic extraction can promote the proliferation and migration of IEC-6cells at certain concentrations.
