摘要
建立了应用超高效液相-高分辨飞行时间质谱联用(Ultra-Performance Liquid Chromatography Time-of-Flight Mass Spectrometry,UPLC-TOF/MS)分析不同产地北五味子中标志性成分的分析方法。采用UPLC-TOF/MS对来源于不同地区的北五味子样品进行分析,通过得到的一级质谱精确质荷比和二级质谱碎片信息,结合软件数据库匹配,对成分进行鉴别。鉴别后采用主成分分析(Principal Component Analysis,PCA)和正交偏最小二乘法-判别分析(Orthogonal Partial Least Squares Discriminant Analysis,OPLS-DA)对得到的数据进行分类和统计学分析,筛选差异组分。经上述方法进行处理和分析后,初步筛选并鉴定出了不同产地北五味子的12种具有统计学差异的组分。本研究建立了UPLC-TOF/MS分析不同产地北五味子的分析方法,建立的方法有效、可靠,可用于不同产地的五味子的有效性成分研究和产地鉴别。
Established a method to screening iconic components in Schisandra chinensis(Turcz.)Baill by Ultra Performance Liquid Chromatography Time-of-Flight Mass Spectrometry.UPLC-TOF/MS was used to determine Schisandra chinensis samples from different regions,through the accurate mass charge ratio of primary mass spectrometry and fragment information of secondary mass spectrometry,combined with software database to identification of iconic components.PCA and OPLS-DA were used for classification,data processing and screening the diversity components by statistical analysis to find the iconic components of Schisandra chinensis(Turcz.)Baill in different regions.After analysis by above mentioned methods,12 different components were obviously distinguished and iconic components with significant differences were screened.In this study,UPLC-TOF/MS was successfully established for the identification of Schisandra chinensis(Turcz.)Baill.in different regions.The method of this experiment is effective and reliable and can be applied for origin identification and distinguish iconic components of Schisandra chinensis(Turcz.)Baill.from different regions.
