高分辨质谱快速筛查药食同源产品中5种真菌毒素

Rapid Determination of Five Mycotoxins in Medicine and Food Homologous Products by Liquid Chromatography-High Resolution Mass Spectrometry

摘要 建立了人参、五味子、刺五加、人参花、灵芝和灵芝孢子粉基质中5种真菌毒素的液相色谱-高分辨质谱检测及筛查方法,样品中残留的真菌毒素采用含1%甲酸的乙腈提取,经氯化钠及硫酸镁盐析分层,提取液经无水硫酸镁、N-丙基乙二胺(PSA)和石墨化炭黑(GCB)吸附剂分散固相萃取净化,用高效液相色谱-高分辨质谱定性。在上述检测方法条件下,所测的5种真菌毒素的定量限为10μg/kg,平均回收率为75-95%,线性范围为10-1000μg/L(R2>0.99)。本研究所建立的方法前处理简单快捷,能够快速筛查出样品中的5种真菌毒素并对目标物进行准确的定性分析,为药食同源特色农产品中的真菌毒素残留的风险评估提供了技术支持。 A liquid chromatography-High resolution mass spectrometry(LC-HRMS)method was developed for the detection and screening of 5 fungal toxins in ginseng,Schisandra chinensis,Acanthopanax senticosus,Ginseng,Ganoderma lucidum and Ganoderma lucidum spore powder.The fungal toxins in the sample were extracted from acetylene containing 1%formic acid,salted out by sodium chloride and magnesium sulfate,and purified by solid phase extraction with anhydrous magnesium sulfate,PSA and GCB.The samples were identified by high performance liquid chromatography high resolution mass spectrometry.Under the above conditions,LOQ of 5 fungal toxins was 10μg/kg.The average recovery was 75%~95%,and the linear range was 10~1000μg/L(R2>0.99).The method established in this study is simple and quick,and can quickly screen 5 mycotoxins in samples and make accurate qualitative analysis of the target substances,it provides technical support for risk assessment of mycotoxin residues in food and drug homology agricultural products.
