摘要
目的观察调经养巢方剂对卵巢早衰(Premature Ovarian Failure,POF)模型大鼠血清性激素、卵巢颗粒细胞(granulosa cells,GCs)超微结构及细胞凋亡的影响。方法将25只SPF级成熟雌性SD大鼠随机分成空白组、模型组、调经养巢膏剂和汤剂组、补佳乐组,每组5只。除空白组自由喂养,不予干预外,其他组均经腹腔注射去氧乙烯基环己烯(VCD)建立POF大鼠模型,灌胃治疗后,通过阴道涂片观察动情周期变化,检测大鼠血清性激素、卵巢GCs超微结构、细胞凋亡水平变化。结果与空白组比较,模型组大鼠动情周期明显延长,血清促性腺激素释放激素(GnRH)、促卵泡生长激素(FSH)、促黄体生成素(LH)水平显著升高(P<0.05),抗缪勒氏管激素(AMH)、雌激素(E;)水平显著降低(P<0.05);大鼠卵巢GCs超微结构被破坏,线粒体减少,嵴减少甚至消失;细胞凋亡指数显著升高(P<0.05)。与模型组比较,调经养巢膏剂组及汤剂组大鼠动情周期明显缩短,血清激素水平明显改善,调经养巢汤剂组对FSH作用更显著,调经养巢汤剂组对AMH作用更显著;卵巢GCs超微结构明显改善,线粒体增多,嵴增多;细胞凋亡指数显著下降(P<0.05)。结论调经养巢方膏剂和汤剂均可以改善POF大鼠血清激素水平及卵巢GCs超微结构,其具体机制可能是通过修复卵巢GCs内线粒体等细胞器的结构与功能,减少GCs凋亡,从而延缓POF进展;但二者作用又有差异,膏剂调节AMH的作用更显著,汤剂调节FSH的作用更显著。
Objective To observe the effect and its possible mechanism of Tiaojing Yangchao Paste and Decoction on premature ovarian failure(POF)rats.Methods POF was induced in rats with deoxyvinylcyclohexene(VCD).SPF female SD rats were randomly divided into five groups:blank group,model group,Tiaojing Yangchao Paste group,and Tiaojing Yangchao Decoction group,progynova group.Serum sex hormone levels,ovarian ultrastructure and cell apoptosis were detected,respectively.Results Compared with blank group,Gn RH,FSH and LH levels were significantly increased(p<0.05),and AMH and estrogen(E;)levels significantly lower(p<0.05)of model group;ovarian granulosa cells(GCs)ultrastructure was damaged;the apoptotic cells were significantly increased.Compared with model group,Gn RH,FSH and LH levels in each Tiaojing Yangchao Paste and Decoction group were significantly decreased(p<0.05),and AMH and E;levels were significantly increased(p<0.05);ovarian GCs ultrastructure were significantly ameliorated,and the mitochondria were significantly increased;the apoptotic degree was significantly lower(p<0.05).Compared Tiaojing Yangchao Paste group with Tiaojing Yangchao Decoction group,FSH level of Tiaojing Yangchao Decoction group was significantly decreased(p<0.05),and AMH level of Tiaojing Yangchao Paste group was significantly increased(p<0.05).Conclusion This study suggested that Tiaojing Yangchao Paste and Decoction could alleviate POF via regulating the structure and function of mitochondrial to inhibit apoptosis;there were differences between Tiaojing Yangchao paste and decoction.The effect on regulating AMH of Tiaojing Yangchao Paste was more significant,while the effect on regulating FSH of Tiaojing Yangchao Decoction was more significant.
