摘要
目的GEPIA数据库分析DKK1在膀胱癌组织中的表达,并探讨与预后的关联。方法GEPIA数据库挖掘DKK1基因在常见肿瘤中的表达量;提取该基因在膀胱癌组织和癌旁的表达差异,分析膀胱癌不同分期患者DKK1的水平;进一步提取该数据库Survival模块数据,研究DKK1基因与膀胱癌患者总体生存率的关系。ELISA法测定膀胱癌和健康体检者血清中DKK1的浓度,并比较其与膀胱癌患者主要临床参数的关联。结果头颈部癌、食管癌、肺癌和胰腺癌组织中DKK1 mRNA表达量明显高于癌旁组织;在膀胱癌和黑色素瘤中则低表达;不同临床分期的膀胱癌组织中DKK1基因表达水平差异无统计学意义(P>0.05);DKK1低表达的膀胱癌组患者总体生存率(OS)高于DKK1高表达组(P<0.05)。膀胱癌患者血清中DKK1浓度显著高于健康体检者的水平,且其浓度值与淋巴结转移和临床分期呈正相关。结论不同肿瘤组织中DKK1表达量不同;膀胱癌组织中DKK1基因明显低表达于癌旁正常组织,且与患者预后相关;血清中DKK1浓度可用来帮助诊断胰腺癌。
Objective To analyze the expression of DKK1 in bladder cancer tissue by GEPIA database and explore its relation with prognosis.Methods GEPIA database was used to detect the expression of DKK1 gene in common tumors.The expression differences of DKK1 gene in bladder cancer tissues and adjacent tissues were extracted to analyze the levels of DKK1 in patients with different stages of bladder cancer.The relationship between DKK1 gene and overall survival rate of patients with bladder cancer was studied by further extraction of survival module data from this database.ELISA method was used to determine the serum DKK1 concentration in bladder cancer patients and healthy subjects,and the correlation between DKK1 concentration and main clinical parameters of bladder cancer patients was compared.Results The expression of DKK1 mRNA in head and neck cancer,esophageal cancer,lung cancer and pancreatic cancer was significantly higher than that in para-cancer tissues.It is low in bladder cancer and melanoma.There was no statistical significance on the difference in DKK1 gene expression in different clinical stages of bladder cancer(P>0.05).The overall survival rate(OS)of patients with low DKK1 expression was higher than that of patients with high DKK1 expression(P<0.05).Serum DKK1 concentration in bladder cancer patients was significantly higher than that in healthy subjects,and its concentration was positively correlated with lymph node metastasis and clinical stage.Conclusion The expression of DKK1 is different in different tumor tissues.The DKK1 gene in bladder carcinoma was significantly lower than normal tissues,and was correlated with the prognosis of the patient.Serum DKK1 concentration can be used to help diagnose pancreatic cancer.
