摘要
目的观察脂多糖(LPS)以及肿瘤坏死因子α(TNF-α)对人鼻黏膜上皮细胞屏障功能的影响。方法选取健康人鼻黏膜上皮细胞(HNEpC)作为研究对象,观察LPS和TNF-α刺激HNEpC 24-48h后7种细胞连接蛋白的mRNA的表达情况以及11种细胞因子的分泌情况。结果实时荧光定量PCR结果显示,LPS和TNF-α刺激HNEpC后,大部分细胞连接蛋白的mRNA表达下调,其中Claudin-4、Claudin-7和ZO-1的mRNA表达下调最为显著;LPS和TNF-α均能显著促进IL-6、IL-8、IL-12p70蛋白的分泌,此外,TNF-α还可以显著促进IL-1β的分泌。结论LPS和TNF-α的刺激可以下调人鼻黏膜上皮细胞的细胞连接蛋白mRNA表达并促进部分细胞因子的分泌,从而对鼻黏膜上皮细胞的物理和免疫屏障功能造成损伤。
Objective The objective of this study was to observe the effects of lipopolysaccharide(LPS)and tumor necrosis factorα(TNF-α)on epithelial cell barrier function of human nasal mucosa.Methods In this study,healthy human nasal epithelial cells(HNEpC)were selected as subjects,and the mRNA expression of 7 conjunction and secretion of 12 cytokines were observed 24-48 hours after LPS and TNF-αstimulation in human nasal epithelial cells.Results Real-time quantitative PCR showed that most conjunction expressions were down-regulated after HNEpC was stimulated by LPS and TNF-α,among which the mRNA expressions of Claudin-4,Claudin-7 and ZO-1 were most significantly down-regulated.Both LPS and TNF-αsignificantly promoted the secretion of IL-6,IL-8,IL-12p70,while TNF-αsignificantly increased secretion of IL-1β.Conclusion LPS and TNF-αcan down-regulate the expression of connexin in human nasal epithelial cells and stimulate the secretion of cytokines,thereby further damaging their physical and immune barrier functions.
