金毛寻回猎犬19个STR基因座的遗传多态性及群体遗传结构分析 被引量:2

摘要 目的我国对犬种属鉴定、个体识别和亲权鉴定的需求日益增加,但对犬种群遗传多样性的研究却很少。本文基于一种商业化犬STR试剂盒来评估生活在我国的金毛寻回猎犬的遗传多态性和遗传结构。方法基于该试剂盒19个犬STR基因座分析了640只金毛寻回猎犬的遗传多态性和法医学应用效能,并基于4个相同的STR基因座将其与匈牙利5个犬类群体、我国的拉布拉多寻回猎犬群进行群体遗传关系分析。结果在经过Bonferroni校正后,这19个STR基因座在金毛寻回猎犬中符合Hardy-Weinberg平衡。19个STR基因座均具有较高的多态性,累积个体识别率为0.999 999 999 999 983 9和累积非父排除率为0.999 511,表明这19个STR基因座可以用于金毛寻回猎犬的个体识别,并能够作为一种辅助工具用于犬类的亲权鉴定。群体遗传结构分析进一步揭示了所研究的金毛寻回猎犬与其他6个参考犬群之间的遗传差异。结论这19个STR基因座在我国金毛寻回猎犬的法医鉴定中具有一定的应用价值,可用于该犬种的个体识别。 Objective The demands for species and individual identification of canines are increasing in China,but the research on genetic diversities of canine breeds is rarely conducted.In this study,we evaluated the genetic polymorphisms and population structure characteristics of the Golden Retriever breed from China.Methods The genetic distributions and forensic efficiencies of 19 canine STR loci in 640 Golden Retriever individuals were analyzed using a commercial canine STR kit,and the genetic relationships among Golden Retriever breed and other 6 reference canine breeds were analyzed based on four overlapped STR loci.Results The results of these 19 STR loci in the Golden Retriever breed were in accordance with the Hardy-Weinberg equilibrium after Bonferroni correction.The 19 STR loci were highly polymorphic.The combined discrimination power was 0.999 999 999 999 983 9 and the cumulative probability of exclusion was 0.999511,indicating that these 19 STR loci could be used for individual identification and used as an auxiliary tool for parentage testing of Golden Retriever breed.Genetic structure analyses among Golden Retriever and other 6 reference canine breeds further revealed the genetic differences between the Golden Retriever breed and other canine breeds.Conclusion These 19 STR loci with relatively high forensic application values could be employed for individual identification of the Golden Retriever breed from China.
