HPLC法测定二氧化氯乳头消毒剂中乳酸的含量

摘要 建立HPLC法测定二氧化氯乳头消毒剂中乳酸的含量并研究此方法的耐用性。采用HPLC法,色谱柱为资生堂Capcell Pak C18 MG(250 mm×4.6 mm,5μm),流动相为0.1%磷酸水溶液-乙腈(97.5∶2.5,V/V),检测波长为215 nm,流速为1 mL/min,柱温为35℃,进样量为20μL。结果表明:乳酸在200~1000μg/mL内线性关系良好(R2=0.9999),检测限为5μg/mL,定量限为13μg/mL,精密度试验峰面积RSD均符合标准,平均回收率为98.50%,供试品工作液在24 h内稳定。耐用性试验结果显示,在测试范围内,检测波长、流速、柱温、流动相比例和色谱柱对样品中乳酸的含量测定均无影响,且分离度符合要求。此方法操作简易,可靠性高,可以用作该乳头消毒剂中乳酸的质量控制。 To establish a method for content determination of lactic acid in chlorine dioxide nipple disinfectant by HPLC,and to study the durability of this method,HPLC method was adopted,the chromatographic column is Shiseido Capcell Pak C18 MG(250 mm×4.6 mm,5μm).The mobile phase was 0.1%phosphoric acid aqueous solution-acetonitrile(97.5∶2.5,V/V).The detection wavelength was 215 nm.The flow rate was 1 mL/min.The column temperature was 35℃.The injection was 20μL.Results showed that lactic acid had a good linear relationship(R2=0.9999)within 200 to 1000μg/mL,the detection limit was 5μg/mL,the quantitative limit was 13μg/mL.Precision testing,all peak area RSD meets the standard.The average recovery rate was 98.50%.A sample working fluid was stably in 24 h.The durability test showed,in the test range,the detection wavelength,flow rate,column temperature,mobile phase ratio and chromatographic column had no effect on the determination of lactic acid content in the sample,and the separation degree meet the requirements.This method was simple and reliability,which can be used as the quality control of lactic acid in the nipple disinfectant.
