青藤碱对IL-1β诱导大鼠滑膜细胞株RSC-364增殖及炎症因子的影响 被引量:1

摘要 目的:观察青藤碱对IL-1β诱导大鼠滑膜细胞株RSC-364增殖及炎症因子的影响。方法:培养大鼠滑膜细胞株RSC-364,进行如下分组,青藤碱不同浓度组(浓度梯度为0、80、160、240、320 mg/L)、IL-1β(10 ng/mL)+青藤碱不同浓度组(浓度梯度为0、80、160、240、320 mg/L)。通过CCK-8方法检测不同浓度青藤碱对大鼠滑膜细胞株RSC-364在IL-1β诱导下增殖率的影响,并通过酶联免疫吸附(ELISA)法检测IL-6、MMP3的含量。结果:青藤碱浓度与大鼠滑膜细胞株RSC-364增殖率呈负相关,浓度增加,增殖率降低,其增殖明显呈剂量依赖性(P<0.05);在10 ng/mL IL-1β诱导下,加入不同浓度青藤碱处理,RSC-364细胞增殖减少(P<0.05),RSC-364细胞炎性因子IL-6、MMP-3的表达下调(P<0.05)。结论:不同浓度青藤碱抑制IL-1β诱导的RSC-364增殖,抑制炎性因子IL-6、MMP-3的分泌,这可能与其在临床中治疗强直性脊柱炎和类风湿关节炎的作用机制有关。 Objective: To observe the effects of sinomenine(SIN) on IL-1β induced proliferation of synovium cell line RSC-364 and inflammatory factors in rats. Methods: Rat synovial cell line RSC-364 was cultured and divided into the sinomenine groups of different concentrations(0 mg/L, 80 mg/L, 160 mg/L, 240 mg/L, 320 mg/L), the IL-1β(10 ng/mL) plus sinomenine group(0 mg/L, 80 mg/L, 160 mg/L, 240 mg/L, 320 mg/L). The proliferation of rat synovial cell line RSC-364 induced by different concentrations of sinomenine was detected by CCK-8 method, and the contents of IL-6 and MMP3 were detected by ELISA. Results: sinomenine concentration was negatively correlated with the proliferation rate of rat synovial cell line RSC-364, and the proliferation rate decreased with the increase of sinomenine concentration(P<0.05). After 10 ng/mL IL-1β induction, the proliferation of RSC-364 cells was decreased(P<0.05), and the expressions of inflammatory factors of IL-6 and MMP-3 in RSC-364 cells were down-regulated(P<0.05). Conclusion: Different concentrations of sinomenine can inhibit the IL-1β induced proliferation of RSC-364 and inhibit the secretion of inflammatory factors of IL-6 and MMP-3, which may be related to its action mechanism in the treatment of ankylosing spondylitis and rheumatoid arthritis in clinic.
